Resumo: | In the domestic rabbit (Oryctolagus cuniculus), the vomeronasal organ ends in the rostral part of the palate through the incisive duct. This duct allows pheromones to enrich and stimulate this organ. The obstruction (cauterization) of the incisive duct causes morphological alteration in the vomeronasal organ and in the testis. In order to develop the present study, incisive ducts of 16 New Zealand male rabbits (60 days of age) were cauterized under anesthesia. Sixteen rabbits from the same breed and age were used as a control group. Hence, 8 animals of each group (control and cauterized) were sacrificed at two different time points: 120 and 180 days of age (d). Before sacrifice, the animals were anesthetized and perfused through the aortic artery with bicarbonate Ringer s solution, followed by a 2.5% glutaraldehyde solution and testes, vomeronasal organ, skeletal muscle and bone were collected. These tissues were submitted to routine procedures for histological studies, transmission electron microscopy, also stereological and morphometrical techniques. Based on the serum testosterone dosages, it was found that the 120d group presented no difference in androgen levels; however, the 180d group showed a variation on levels of serum testosterone. Hence, in order to better understand the results, the 180d group was subdivided in low (180d-LT) and high (180d-HT) testosterone concentration. Apparently, the cauterization procedure did not affect the volume density of the components as well as the epithelium height of vomeronasal organ. However there are some evidence of neuronal loss, based on spaces in the apical portion the sensorial epithelium and elongated, dense, and irregular borders of nuclei of the supporting cells. The tubular diameter was lower in 120d and 180d-HT when compared with the control group. The In comparison to the control, Leydig cell number was higher in the 180d-LT and lower in the 120d group. Also, the volume densities of the testicular parameters were lower in the 120d when compared with control. Regarding to Sertoli cell, it was observed a decrease in the percentage occupied in the testis parenchyma in 120d and 180d-LT groups. . Considering the muscle skeletal system, there are no differences in the bone density of the femoral epiphyses and also in the mass and composition of the femoral muscle. These results demonstrate that much about the VNO remains to be elucidated in the different species of mammals, in which the VNO can act in a different form, as in the pig, which pheromones are essential for sexual stimulation of both males and females. |

Este ducto permite que os feromônios alcancem e estimulem este órgão. A obstrução do ducto incisivo, induzida pela cauterização, ocasiona alterações morfológicas no órgão vomeronasal e no testículo. Para desenvolver o presente estudo, 16 coelhos da raça Nova Zelândia machos sadios, aos 60 dias de idade, tiveram seus ductos incisivos cauterizados após anestesia. Outros 16 coelhos da mesma raça e idade foram utilizados como grupo controle. Posteriormente, oito animais de cada grupo (controle e cauterizado) foram sacrificados em dois momentos diferentes: aos 120 e 180 dias de idade. Para a retirada do órgão vomeronasal, testículos, músculo esquelético e osso, os animais também foram anestesiados e a seguir, foram perfundidos com solução fixadora. Os órgãos foram coletados e submetidos a procedimentos de rotina para estudos histológicos, ultraestruturais e por meio de técnicas estereológicas e morfométricas. Para estudos biométricos, morfometria testicular e dosagens de testosterona, os animais com 180 dias foram divididos em subgrupos com baixas e altas concentrações de testosterona, devido às variações nas concentrações desse hormônio. Os animais com 120 dias não apresentaram esse comportamento. Apesar de não haver diferenças importantes nos percentuais dos componentes do órgão vomeronasal e nos valores de altura do epitélio sensitivo, observou-se na histologia que houve morte de neurônios evidenciados por espaços na porção apical do epitélio sensorial e núcleos das células de sustentação alongados, densos e com bordas irregulares. A morfometria testicular apontou que houve alteração no diâmetro tubular nos animais do grupo tratado com 120 dias e nos animais do subgrupo com altas concentrações de testosterona, onde foi menor, em relação aos animais do grupo controle. Em relação ao número de células de Leydig nos animais adultos tratados do subgrupo com baixa testosterona, os animais tratados tiveram valores maiores, mas nos animais jovens tratados os parâmetros volumétricos da célula e o número de células foi menor em relação aos animais do grupo controle. Os animais tratados dos grupos de 120 dias e de 180 dias com baixa testosterona apresentaram diminuição no percentual ocupado pelas células de Sertoli no parênquima testicular. Os animais tratados com 120 dias e os animais do subgrupo com baixa testosterona tiveram suas concentrações de testosterona sérica menores. O tratamento não afetou a densidade óssea da epífise do fêmur, nem a massa e a composição do músculo reto femoral. Os resultados do presente estudo abrem portas para novas áreas do conhecimento da morfofisiologia da reprodução, uma vez que indica que a cauterização do ducto incisivo atrasa a puberdade e afeta a espermatogênese.
