Estudos mostram que em reações inflamatórias ocorre o aumento da produção de espécies reativas de oxigênio. Dentre as várias espécies reativas nosso estudo analisou a produção de oxigênio singleto (1O2) durante a febre produzida pela administração de LPS. Neste trabalho foram utilizados Ratos Wistar fêmeas que foram submetidas à cirurgia de implante de transmissores de temperatura na cavidade intraperitoneal. Os animais receberam injeção endovenosa de LPS (5 μg/kg) ou seu veículo (salina 0,9%, 2 mL/Kg) e tiveram a temperatura corporal aferida a cada 10 minutos, durante 1h, 2,5h ou 6h após a injeção. Em seguida, os animais foram eutanasiados para a dissecação do hipotálamo e tecido adiposo marrom. A concentração de 1O2 foi investigada por ressonância paramagnética eletrônica nesses tecidos utilizando-se o marcador de TEMP. Os resultados mostraram que há um aumento significativo na detecção de 1O2 tanto no hipotálamo quanto no tecido adiposo marrom em animais do grupo LPS comparado com o grupo controle 2,5 h após a administração. Nos grupos de 1 h e 6 h apesar de haver uma tendência de aumento desta espécie reativa, não foram encontradas diferenças significativas. A avaliação temporal da concentração de 1O2 revelou que o pico de aumento dessa espécie reativa no hipotálamo e tecido adiposo marrom coincidiu com o pico de elevação da temperatura corporal, o que sugere que as vias de produção de 1O2 são ativadas e que podem ter papel relevante na patogênese da febre. __________________________________________________________________________ ABSTRACTFever is the increasing of the body temperature for an acute phase inflammatory reaction. Studies have shown that in inflammatory reactions there is an increasing of the production of oxygen reactive species. Among the various reactive species, our study has assessed the production of singlet oxygen (1O2) during fever induced by the administration of LPS. This work was carried out with female Wistar rats that underwent a temperature transmitter’s implant surgery in the intraperitoneal cavity. The animals received an intravenous injection of LPS (5 μg/kg) or its control (saline 0,9%, 2 mL/Kg), and had its body temperature measured every 10 minutes, during 1h, 2.5h, or 6h after the injection. After that, the animals were euthanized for the dissection of the hypothalamus and the brown adipose tissue. The 1O2 concentration was assessed by electron paramagnetic resonance in these tissues, using the TEMP’s marker. Results showed a significant increasing in the detection of 1O2 both on hypothalamus and brown adipose tissue in animals of the LPS group, compared to the control group 2.5h after the administration. In groups 1h and 6h, despite the rising trend of this reactive specie, there was no significant differences found.
