PHYTOCHEMISTRY, TOXICOLOGICAL AND MICROBIOLOGICAL STUDIES OF Costus spicatus JACQ. LEAVES

Resumo: The vegetal speciel Costus spicatus Jacq., called “cana-do-brejo” in Brazil, belong to Zingiberaceae family, and it’s used in the popular medicine, especially in the Amazon region as a depurative and diuretic, relieving urinary tract infections and aiding in the removal of kidney stones. The objetictive of this study was perform the phytochemical study in search of main classes of secondary metabolities, microbial activity and cytotoxicity with the Brine Shrimp Test of ethanolic crude extract of dried leaves of vegetal specie C. spicatus. The phytochemical Trial was realized was achieved by chemical methods to determine the following metabolites: alkaloids, phenols and tannins, flavonoids, anthraquinones, saponins, triterpenes, reducing sugars, polysaccharides, organic acids and steroids. The toxicity bioassay with Brine Shrimp Test (BST) was performed the test with Artemia salina, which was considered the death or life of nauplius. In microbiological test was used the diffusion with standard bacterial strains with concentrations of 25 µg/mL, 50 µg/mL and 100 µg/mL of EBE of C. spicatus. In this perspective, in this study we observed in EBE the presence of alkaloids, and too, phenols and tannins. The extract of C. spicatus showed a high degree of toxicity in concentrations analyzed with BST. To estimate the presence of antimicrobial activity was measured the size of the circles of inhibition of the extract ant antibiotics, verifying that after 24 hours of incubation there was no significant growth of inhibitory circles, where it was determined that there is no inhibitory effect in C. spicatusfront of the bacterial stains used and concentrations used.Keywords: Costus spicatus; phytochemical screening; toxicity bioassay; Artemia salina; microbiological test.

(cana-do-brejo), espécie pertencente à família Zingiberaceae, é utilizada na medicina popular, principalmente na região amazônica como depurativa e diurética, aliviando infecções urinárias e auxiliando na eliminação de pedras renais. O objetivo desta pesquisa foi realizar o estudo fitoquímico em busca das principais classes de metabólitos secundários, atividade microbiológica e citotóxica frente a Artemia salina do extrato bruto etanóico das folhas de Costus spicatus. O teste fitoquímico foi realizado por métodos químicos, visando determinar os seguintes metabólitos: alcalóides, fenóis e taninos, flavonóides, antraquinonas, saponinas, triterpenos, açucares redutores, polissacarídeos, ácidos orgânicos e esteróides. Quanto ao bioensaio de citotoxicidade foi realizado o teste com Artemia salina, onde se considerou a morte ou vida dos metanáuplios. O teste microbiológico foi utilizado a prática de difusão em disco com linhagens bacterianas padronizadas em concentrações de 25μg/mL, 50μg/mL e 100μg/mL do extrato bruto etanólico. Nessa perspectiva, foi possível observar a presença de alcaloides, e também, fenóis e taninos. O extrato bruto apresentou alto grau de toxicidade nas concentrações A. salina.
