Morphoanatomical study, chemical and biological analysis of the methanol extract of the leaves of Mabea angustifolia Spruce ex Benth. (Euphorbiaceae)

Resumo: This study aimed to evaluate the chemical profile of the methanol extract (EMA-Met) from the leaves of Mabea angustifolia Spruce ex Benth., its biological and describe its morphoanatomy. M. angustifolia leaves were collected in the municipality of Itacoatiara, Amazonas, Brazil. After, they were dried, ground and subjected to extraction with solvents in increasing order of polarity. The EMA-Met extract was submitted to mass spectrometry analysis (APCI-MS) and evaluated by phytochemical screening. The biological action of the extracts was tested on Candida albicans (CP Robin) Berkhout 1923, Candida parapsilosis (Ashford) Langeron & Talice 1932 and on Salmonella Lignieres 1900 spp., Escherichia coli (Migula 1895) and Staphylococcus aureus Rosenbach 1884 bacteria. The morphoanatomical analysis of the leaves identified characteristics of diagnostic value for the species, such as leaf shape and margin, number and position of nectaries and colleters, branched tector trichomes and papillose cells on the abaxial surface of the epidermis. The phytochemical analysis revealed the presence of saponins, condensed tannins, phenols and flavonoids. The analysis by APCI-MS allowed the identification flavonoids of recognized biological activity in the literature, being two O-glycosides of quercetin, 3-O-α-L-arabinopirasil quercetin and quercetin-3-O-glucuronide, in addition to quercetin and myricetin-3-O-ramanoside. The extract solution at a concentration of 10 mg/mL inhibited E. coli, with an average halo diameter of 5 mm, the inhibition halo for C. albicans presented a diameter mean of 15 mm and for C. parapsilosis it was 16 mm. The results are promising, showing the antimicrobial potential of M. angustifolia.

Se recolectaron hojas de M. angustifolia en municipio de Itacoatiara, Amazonas, Brasil. En seguida, se secaron, se trituraron y se sometieron a extracción con solventes en orden creciente de polaridad. El extracto de EMA-Met se sometió a análisis de espectrometría de masas (APCI-MS) y se evaluó mediante cribado fitoquímico. La acción biológica de los extractos fue probada en Candida albicans (CP Robin) Berkhout 1923, Candida parapsilosis (Ashford) Langeron & Talice 1932 y en Salmonella Lignieres 1900 spp., Escherichia coli (Migula 1895) y Staphylococcus aureus Rosenbach 1884 bacterias. La morfoantomía foliar identificaron características valiosas para la identificación de la especies, tales como forma y margen de la hoja, número y posición de nectarios y coléteres, tricomas tectores ramificados y células papilares en la superficie abaxial de la epidermis. El análisis fitoquímico reveló la presencia de saponinas, taninos condensados, fenoles y flavonoides. El análisis por APCI-MS permitió identificar compuestos flavonoides de reconocida actividad biológica en la literatura, siendo dos O-glucósidos de quercetina, 3-O-α-L-arabinopirasil quercetina y quercetina-3-O- glucurónido, además de quercetina y miricetina-3-O-ramanoside. Se utilizó la técnica del pozo para verificar la actividad antimicrobiana de la muestra, la solución de extracto a una concentración de 10 mg / mL inhibió a E. coli, con un halo de diámetro promedio de 5 mm, el halo de inhibición para C. albicans presentó un diámetro promedio de 15 mm y para C. parapsilosis fue de 16 mm. Los resultados son prometedores y muestran el potencial antimicrobiano de M. angustifolia.Este estudo teve como objetivo avaliar o perfil químico do extrato metanólico (EMA-Met) das folhas de Mabea angustifolia Spruce ex Benth., sua ação biológica e descrever sua morfoanatomia. As folhas de M. angustifolia foram coletadas no município de Itacoatiara, Amazonas, Brasil. Em seguida, foram secas, trituradas e submetidas à extração com solventes em ordem crescente de polaridade. A análise morfoanatômica das folhas identificou características valiosas para a identificação da espécie, como a forma e margem foliar, número e posição de nectários e coléteres, tricomas tectores ramificados e células papilosas na face abaxial da epiderme. A análise fitoquímica revelou a presença de saponinas, taninos condensados, fenóis e flavonoides. A análise por APCI– MS permitiu identificar flavonoides de reconhecida atividade biológica na literatura, sendo dois O- glicosídeos da quercetina, o 3-O-α-L-arabinopirasil quercetina e a quercetina-3-O-glucuronídeo, além da quercetina e a miricetina-3-O-ramanosídeo. A solução do extrato na concentração de 10 mg/mL inibiu a E. coli, com diâmetro médio do halo de 5 mm, o halo de inibição para C. albicans apresentou diâmetro médio de 15 mm e para C. parapsilosis foi de 16 mm.
