Use of proteins associated with pregnancy (PAG’s - Bovine Pregnancy Test Idexx®) as a method of early diagnosis of pregnancy in gyrolando dairy cows

Resumo: The objective of this study was to observe the effectiveness of the use of proteins associated with pregnancy (PAG’s) as a method of early pregnancy diagnosis in Girolando dairy cows. By using a colorimetric reaction test, recommended for use in cows from the 28th day after service, VISUAL-PAG from "IDEXX Laboratories". The sensitivity and specificity of the test were assessed at 25, 26, 27 and 28 days post-TAI, using 57 multiparous females submitted to four sequential collections of blood samples on the days described. The material was collected by venipuncture of the coccidian vessels in vacuum tubes without anti-coagulant, processed and stored at -20ºC for later analysis. At 32 days of TAI all females were subjected to the diagnosis of pregnancy by ultrasonography (US), the Gold Test of the experiment. The results were obtained through a concordance study, which revealed 100% sensitivity and 94.7% specificity for days 25, 26, 27 and 28 post-TAI. In view of these results, it can be considered that the VISUAL-PAG kit can be used as an early and safe method of early pregnancy diagnosis in Girolando dairy cows at 25 days after artificial insemination.

Através de una prueba de reacción colorimétrica, recomendada para su uso en vacas a partir del 28° día posterior al servicio, el VISUAL-PAG de "IDEXX Laboratories". La sensibilidad y especificidad de la prueba se verificaron a los 25, 26, 27 y 28 días después de la Inseminación Artificial de Tiempo Fijo (IATF), utilizando 57 hembras multíparas, enviadas a cuatro colecciones secuenciales de muestras de sangre en los días descritos. El material se recolectó mediante venopunción de los vasos coccígeos en tubos de vacío sin anticoagulante, se procesó y almacenó a -20ºC para su posterior análisis. A los 32 días después de IATF, todas las hembras fueron sometidas a un diagnóstico de embarazo por ultrasonido (US), la prueba estándar del experimento. Los resultados se obtuvieron a través de un estudio de acuerdo, donde reveló un 100% de sensibilidad y un 94,7% de especificidad para los días 25, 26, 27 y 28 posteriores a la IATF. En vista de estos resultados, se puede considerar que el kit VISUAL-PAG puede usarse como un método de diagnóstico precoz y seguro del prenhez en vacas lecheras Girolando a los 25 días después de la inseminación artificial.Objetivou-se com este estudo observar a eficácia da utilização das Proteínas Associadas à Gestação (PAG´s) como método de diagnóstico precoce de gestação em vacas leiteiras da raça Girolando. Por meio de um teste de reação colorimétrica, recomendado para uso em vacas a partir do 28º dia pós-serviço, o VISUAL-PAG do “IDEXX Laboratories”. Verificou-se a sensibilidade e especificidade do teste aos 25, 26, 27 e 28 dias pós-Inseminação Artificial em Tempo Fixo (IATF), utilizando-se 57 fêmeas multíparas, submetidas a quatro coletas sequenciais de amostras de sangue nos dias descritos. O material foi coletado através de venopunção dos vasos coccígenos em tubos de vácuo sem anticoagulante, processado e armazenado a -20ºC para posterior análise. Aos 32 dias da IATF todas as fêmeas foram submetidas ao diagnóstico de gestação por ultrassonografia (US), teste padrão do experimento. Os resultados foram obtidos através de um estudo de concordância, onde revelou sensibilidade de 100% e especificidade de 94,7% iguais para os dias 25, 26, 27 e 28 pós-IATF.
