Artificial insemination in quails through different methods of semen collection

Resumo: The objective of this study was to evaluate different methods of semen collection in quails and the possible viability of artificial insemination. Two collection methods were used: method 1: Abdominal massage in the male, and method 2: Abdominal massage in the female after male copulation. Were used 16 females and 13 males of the species Coturnix coturnix coturnix, aged 10 weeks. In the first method, the selected male was stimulated with the abdominal massage technique to release a foamy secretion through the glands for cloacal and his reproductive apparatus, which was collected in a disposable 1 ml insulin syringe, immediately and disposed in the vagina region of each pair of females in method 1. In the second method 2, the copulation of the selected male was performed in the mannequin females which received an abdominal massage for the removal of the male ejaculate. The eggs of each method were marked and incubated for 18 days. There was a fertility index of 0% in method 1, and 4.88% in method 2. After failure in fertility and feasibility rates, tests for sperm detection were sought in the male reproductive system through histological cuts. The conclusion was that the methods of artificial insemination were not efficient because did not exert the necessary pressure to enable sperm. Thus, the methods of semen collection and artificial insemination applied obtained undesirable indices and are not viable for reproductive management in quails.

Se utilizaron dos métodos de colección: método 1: Masaje abdominal en el macho, y método 2: Masaje abdominal en la hembra después de la cópula masculina. Utilizamos 16 hembras y 13 machos de la especie Coturnix coturnix coturnix, con 10 semanas de edad. En el primer método, el macho seleccionado fue estimulado con la técnica de masaje abdominal, haciendo que liberara una secreción espumosa a través de las glándulas para los cloácales y su aparato reproductivo, que se recogió en una jeringa desechable de 1 ml de insulina, siendo posteriormente aplicado inmediatamente en la región vagina de cada par de hembras en el método 1. En el segundo método 2, la cópula del macho seleccionado se realizó en las hembras de maniquí, donde recibieron un masaje abdominal para la extracción de la eyaculación masculina. Los huevos de cada método fueron marcados e incubados durante 18 días. Hubo un índice de fertilidad del 0% en el método 1, y del 4,88% en el método 2. Después de fallas en las tasas de fertilidad y viabilidad, se buscaron pruebas para la detección de espermatozoides en el sistema reproductivo masculino a través de cortes histológicos. Se llegó a la conclusión preverque los métodos de inseminación artificial no eran eficientes porque no ejercían la presión necesaria para permitir los espermatozoides. A partir de cortes histológicos fue posible identificar y localizar espermatozoides en todo el sistema reproductivo en el macho específicamente en el conducto deferente.  Por lo tanto, los métodos de recolección de semen e inseminación artificial aplicados obtuvieron índices indeseables, por lo que no son métodos viables de gestión reproductiva en las codornices.Objetivou-se avaliar diferentes métodos de coleta de sêmen em codornas e a possível viabilidade da inseminação artificial. Foram utilizados dois métodos de coleta: método 1: Massagem abdominal no macho, e método 2: Massagem abdominal na fêmea após a cópula do macho. Foram utilizadas 16 fêmeas e 13 machos da espécie Coturnix coturnix coturnix, com idade de 10 semanas. No primeiro método, o macho selecionado foi estimulado com a técnica massagem abdominal, fazendo-o liberar uma secreção espumosa pelas glândulas para cloacais e seu aparelho reprodutivo, a qual era coletada em uma seringa descartável de insulina de 1 ml, sendo posteriormente aplicada imediatamente na região da vagina de cada dupla de fêmeas do método 1. Já no segundo método 2, foi realizada a cópula do macho selecionado nas fêmeas manequins, onde as mesmas receberam uma massagem abdominal para a retirada do ejaculado do macho. Os ovos de cada método foram marcados e incubados durante 18 dias. Houve um índice de fertilidade de 0% no método 1, e de 4.88% no método 2. Após o insucesso nas taxas de fertilidade e viabilidade, procuraram-se os testes para detecção dos espermatozoides no sistema reprodutivo do macho através de cortes histológicos. Chegou-se à conclusão previa os métodos de inseminação artificial não foram eficientes por não exercerem a pressão necessária para viabilizar o espermatozoide.
