The cooperation between genetic and epigenetic factors in the pathogenesis of Alzheimer's disease

Resumo: Aging is an inevitable result of life, characterized by a progressive decline in the function of tissues and organs and an increased risk of mortality. Along with the increase in the elderly population, observed in recent years, there is an increase in the prevalence of neurodegenerative diseases, such as Alzheimer's Disease (AD). The present study consists of an integrative literature review in order to identify the influence of genetic and epigenetic factors in the pathogenesis of AD. The search was carried out during the months of September, October and November 2022, considering studies published in the period between 2014 and 2022, in the following databases: VHL, PubMed MEDLINE, SCIELO, Google Scholar and EbscoHost. It is now very clear that all characteristics of an individual are not determined by DNA alone, but by environment, stress, lifestyle and nutrition that play a vital role in determining an organism's response. In this sense, epigenetic mechanisms involve structural and biochemical changes in chromatin, without alterations in the DNA sequence, which disfigure the expression pattern of genes, affecting several physiological and pathological processes. Therefore, this imbalance in the homeostasis of epigenetic mechanisms is the basis for the abnormal expression of genes related to synaptic plasticity and memory, areas that are notably altered in the initial manifestations of AD. Given the reversible nature of epigenetic mechanisms, these pathways provide promising avenues for therapy against cognitive decline and better prognosis for the disease.

Junto al aumento de la población anciana, observado en los últimos años, se produce un aumento de la prevalencia de enfermedades neurodegenerativas, como la Enfermedad de Alzheimer (EA). El presente estudio consiste en una revisión integrativa de la literatura con el fin de identificar la influencia de factores genéticos y epigenéticos en la patogenia de la EA. La búsqueda se realizó durante los meses de septiembre, octubre y noviembre de 2022, considerando estudios publicados en el período comprendido entre 2014 y 2022, en las siguientes bases de datos: BVS, PubMed MEDLINE, SCIELO, Google Scholar y EbscoHost. Ahora está muy claro que todas las características de un individuo no están determinadas únicamente por el ADN, sino por el medio ambiente, el estrés, el estilo de vida y la nutrición que juegan un papel vital en la determinación de la respuesta de un organismo. En este sentido, los mecanismos epigenéticos involucran cambios estructurales y bioquímicos en la cromatina, sin alteraciones en la secuencia del ADN, que desfiguran el patrón de expresión de los genes, afectando diversos procesos fisiológicos y patológicos. Por tanto, este desequilibrio en la homeostasis de los mecanismos epigenéticos es la base de la expresión anormal de genes relacionados con la plasticidad sináptica y la memoria, áreas que se encuentran notablemente alteradas en las manifestaciones iniciales de la EA. Dada la naturaleza reversible de los mecanismos epigenéticos, estas vías brindan vías prometedoras para la terapia contra el deterioro cognitivo y un mejor pronóstico para la enfermedad.O envelhecimento é um resultado inevitável da vida, caracterizado pelo declínio progressivo da função dos tecidos, órgãos e elevação do risco de mortalidade. O presente estudo consiste em uma revisão integrativa da literatura com o intuito de identificar a influência de fatores genéticos e epigenéticos na patogênese da DA. A busca foi realizada durante os meses de setembro, outubro e novembro de 2022, considerando-se estudos publicados no período compreendido entre 2014 e 2022, nas seguintes bases de dados: BVS, PubMed MEDLINE, SCIELO, Google Scholar e EbscoHost. Atualmente, é muito claro que todas as características de um indivíduo não são determinadas apenas pelo DNA, mas pelo ambiente, estresse, estilo de vida e nutrição que desempenham papel vital na determinação da resposta de um organismo. Nesse sentido, mecanismos epigenéticos envolvem modificações estruturais e bioquímicas na cromatina, sem alterações na sequência de DNA, que desconfiguram o padrão de expressão de genes, afetando vários processos fisiológicos e patológicos. Logo, esse desequilíbrio na homeostase dos mecanismos epigenéticos são a base da expressão anormal de genes relacionados à plasticidade sináptica e à memória, áreas notadamente alteradas nas manifestações iniciais da DA.
