Comparative study of cryopreservation techniques in somatic tissue of Felis silvestris catus

Resumo: The aim of this study was to compare the effects of Slow Freezing (SF) and Solid-Surface Vitrification (SSV) on the cryopreservation of the apical ear skin of domestic cats using histological tools. Fragments of skin obtained from six cats (Felis silvestres catus) were processed and destined for cryopreservation by SF and SSV, using a solution composed of Dulbecco’s Modified Eagle Medium (DMEM) plus dimethyl sulfoxide (DMSO) at 1.5 M or 3.0 M, for SF and SSV, respectively, in addition to 0.25 M sucrose and 10% fetal bovine serum (FBS). After warming, the samples were destined for histological processing for subsequent staining with hematoxylin-eosin, Gromori trichrome, and Weigert staining, as well as the marking of nucleolus organizing regions (NORs) using silver nitrate. Thus, using the VSS technique, characteristics such as epidermis thickness, dermis, and total skin thickness, presented values similar to the control group (p > 0.05). The VSS technique proved to be efficient in maintaining the number of epidermal cells and the percentage of collagen fibers. It is concluded that the application of the VSS technique proved to be superior in the histological maintenance of the apical auricular skin of domestic cats, assisting in the generation of a somatic bank in the species.

Fragmentos de piel obtenidos de seis gatos (Felis silvestres catus)  fueron procesados ​​y destinados a la crioconservación por CL y VSS, utilizando una solución compuesta de Medio Esencial Mínimo Modificado por Dulbecco (DMEM) más dimetil sulfóxido (DMSO) a 1,5 M o 3,0 M, para CL y VSS, respectivamente, además de 0,25 M sacarosa y 10% de suero fetal bovino (SFB). Después del calentamiento, las muestras se destinaron al procesamiento histológico para la tinción posterior con tinción con hematoxilina-eosina, tricromo gromico y coloración de Weigert, así como el marcado de regiones organizadoras de nucleolos (NORs) utilizando nitrato de plata. Por lo tanto, utilizando la técnica VSS, características como el grosor de la epidermis, la dermis y el grosor total de la piel, presentaron valores similares al grupo de control (p > 0,05). La técnica VSS demostró ser eficiente para mantener el número de células epidérmicas y el porcentaje de fibras de colágeno. Resulta que la aplicación de la técnica VSS demostró ser superior en el mantenimiento histológico de la piel auricular apical de los gatos domésticos, ayudando en la generación de un banco somático en la especie.O objetivo do presente trabalho foi comparar os efeitos da Congelação Lenta (CL) e da Vitrificação em Superfície Sólida (VSS) na criopreservação da pele auricular apical de gatos domésticos utilizando ferramentas histológicas. Fragmentos de pele obtidos a partir de seis gatos (Felis silvestres catus) foram processados e destinadas à criopreservação por CL e VSS, utilizando uma solução composta por Meio Essencial Mínimo Modificado por Dulbecco (DMEM) acrescida de dimetilsulfóxido (DMSO) a 1,5 M ou 3,0 M, para CL e VSS, respectivamente, além de 0,25 M de sacarose e 10% de soro fetal bovino (SFB). Após o aquecimento, as amostras foram destinadas ao processamento histológico para posterior realização das colorações com hematoxilina-eosina, tricrômico de Gômori, e coloração de Weigert, bem como foi realizada a marcação de regiões organizadoras de nucléolo (NORs) utilizando nitrato de prata. Na técnica de VSS, características como espessura da epiderme, derme e espessura total da pele, apresentaram valores similares ao grupo controle (p > 0,05). Além disso, a VSS mostrou-se eficiente na manutenção do número de células epidermais e percentual de fibras de colágenas.
