Berberine: chemical, pharmacological aspects and its potential prophylactic relationship with Alzheimer's disease

Resumo: | This article aimed to review the chemical and pharmacological aspects of berberine, correlating them with the pathophysiological outcome of Alzheimer's disease (AD). A descriptive study of the narrative literature review type was carried out. The investigative locus was carried out through an electronic search in the Scientific Electronic Library (SciELO), Latin American and Caribbean Center on Health Sciences Information (BIREME-PAHO-WHO) and National Library of Medicine, USA (PubMed), being Articles published between 2018 and 2023 in English, Portuguese and Spanish were considered. The search for articles was based on the search for the keywords berberine and Alzheimer's disease, using the Boolean operator and to connect these terms. Berberine is classified as a quaternary ammonium salt, it is part of the group of isoquinoline alkaloids called protoberberine. Some studies indicate that berberine is able to attenuate the inflammatory process, excitotoxicity, excessive activity of free radicals and deposition of senile plaques. This review summarized the chemical and physical-chemical aspects of berberine, relating them to bioavailability parameters; clarified some points of the etiology, pathophysiology and clinical manifestations of Alzheimer's disease, considering its complexity; and correlated them considering the main action switches of the neuroprotective substance. In vivo and in vitro studies demonstrate that berberine has therapeutic potential for the treatment of Alzheimer's disease. However, it is still not possible to state whether these results are translational. To verify its effectiveness in humans and estimate possible therapeutic doses, if necessary, robust clinical studies. |

Se realizó un estudio descriptivo del tipo revisión bibliográfica narrativa. El locus investigativo se realizó a través de una búsqueda electrónica en la Biblioteca Científica Electrónica (SciELO), Centro Latinoamericano y del Caribe de Información en Ciencias de la Salud (BIREME-OPS-OMS) y Biblioteca Nacional de Medicina, USA (PubMed), encontrándose artículos publicados entre Se consideraron los años 2018 y 2023 en inglés, portugués y español. La búsqueda de artículos se basó en la búsqueda de las palabras clave berberina y enfermedad de Alzheimer, utilizando el operador booleano y para conectar estos términos. La berberina se clasifica como una sal de amonio cuaternario, forma parte del grupo de alcaloides de isoquinolina denominados protoberberina. Algunos estudios indican que la berberina es capaz de atenuar el proceso inflamatorio, la excitotoxicidad, la actividad excesiva de los radicales libres y el depósito de placas seniles. Esta revisión resumió los aspectos químicos y físico-químicos de la berberina, relacionándolos con parámetros de biodisponibilidad; aclaró algunos puntos de la etiología, fisiopatología y manifestaciones clínicas de la enfermedad de Alzheimer, considerando su complejidad; y los correlacionó considerando los principales interruptores de acción de la sustancia neuroprotectora. Los estudios in vivo e in vitro demuestran que la berberina tiene potencial terapéutico para el tratamiento de la enfermedad de Alzheimer. Sin embargo, todavía no es posible afirmar si estos resultados son traslacionales. Para verificar su eficacia en humanos y estimar posibles dosis terapéuticas, si es necesario, estudios clínicos robustos.Este artigo objetivou revisar os aspectos químicos e farmacológicos da berberina, correlacionando-os com a desfecho fisiopatológico da doença de Alzheimer (DA). Foi realizado um estudo descritivo do tipo revisão narrativa de literatura. O lócus investigativo se deu mediante busca eletrônica nas bases Scientific Eletronic Library (SciELO), Centro Latino-americano e do Caribe de Informação em Ciências da Saúde (BIREME-OPAS-OMS) e National Library of Medicine, dos EUA (PubMed), sendo considerados os artigos publicados entre 2018 e 2023, no idioma inglês, português e espanhol. A busca dos artigos se deu a partir da pesquisa das palavras-chave berberine e Alzheimer disease, usando-se o operador booleano and para conectar esses termos. A berberina é classificada como um sal de amônio quaternário, faz parte do grupo de alcaloides isoquinolínicios denominado protoberberina. Alguns estudos apontam que a berberina é capaz de atenuar o processo inflamatório, a excitotoxicidade, a atividade excessiva de radicais livres e deposição de placas senis. Esta revisão resumiu os aspectos químicos e físico-químicos da berberina, relacionando-os com parâmetros de biodisponibilidade; elucidou alguns pontos da etiologia, da fisiopatologia e das manifestações clínicas da doença de Alzheimer, considerando sua complexidade; e correlacionou-os considerando os principais mecanismos de ação da neuroprotetora substância. Os estudos in vivo e in vitro demonstram que a berberina possui potencial terapêutico para o tratamento da doença de Alzheimer. Entretanto, ainda não é possível afirmar se esses resultados são translacionais.
