Post-mortem muscle pH curve in rats: a preliminary study

Resumo: Rat as experimental model is a highly common practice in many areas of science. However, are not yet available studies using rats as a model for detecting changes in transformation of the muscle in meat. Thus, the objectives of this study were: (1) to describe and standardize the pH curve in transformation into meat of the muscles Triceps brachii, Longissimus dorsi and Biceps femoralis of rats subjected to normal breeding conditions from euthanasia to 24 hours post-mortem; and (2) to determine whether the stress by rats restraining in the ante-mortem period cause qualitative changes in meat and affects the time and pH values for the transformation of muscle in meat. For the determination of muscle pH curves were used 12 male Wistar rats. The animals were euthanized and the carcasses were placed under refrigeration for measurements of muscle pH. Another group of twelve animals was subjected to stress by restraining and after was performed the euthanasia and pH analysis. The results indicate that the values of pH post-mortem in the rat muscle showed similar decline according to commercial species. The muscle pH averages differ significantly from time 0 to 6 hours after slaughter (p<0.05), when the minimum value of muscle pH is found. Rats in restraint stress showed a decline in muscle pH significantly faster than normal curve (p<0.05). Rats are a promising experimental model to detect muscle post-mortem changes, because the decrease in the muscle pH follows the values and time similar to found in the commercial species.

Sin embargo, aún no se dispone de estudios que utilicen ratas como modelo animal en la transformación de músculo en carne. Los objetivos de este estudio fueron: (1) estandarizar la curva de pH de la transformación de la carne de los músculos Triceps brachii, Longissimus dorsi y Biceps femoralis de ratas sometidas a condiciones normales de crianza desde la eutanasia hasta las 24 horas post-mortem; y (2) determinar si el estrés causado por la contención de las ratas en el período ante-mortem interfiere con la curva de pH normal de la transformación de músculo en carne en las ratas. Para determinar la curva de pH muscular, se utilizaron 12 ratas Wistar macho. Los animales se sacrificaron y las canales se colocaron a 4 ° C para la medición del pH muscular. Otro grupo de 12 animales fue sometido a estrés mediante inmovilización durante una hora. Después de este período, los animales fueron sacrificados y se realizaron evaluaciones de pH. Los resultados indicaron que la curva de pH muscular post-mortem de las ratas mostró una tasa de caída similar a la descrita para otras especies de carnicería. Los promedios de pH muscular muestran diferencias significativas desde la hora 0 a las 6 horas después de la eutanasia (p <0.05), cuando se encontró el valor mínimo de pH muscular. Las ratas en condiciones de estrés de contención mostraron una disminución en el pH del músculo significativamente más rápido que la curva normal (p <0.05). Así, se concluye que el uso de ratas como modelo animal para la detección de cambios musculares post-mortem es prometedor, ya que el pH muscular sigue los valores y el tiempo de caída similares a las especies comerciales.A utilização de ratos como modelo experimental é uma prática recorrente em várias áreas da ciência. No entanto, ainda não estão disponiveis trabalhos que utilizem os ratos como modelo animal na transformação de músculo em carne. Os objetivos deste estudo foram: (1) padronizar a curva de pH da transformação em carne dos músculos Triceps brachii, Longissimus dorsi e Biceps femoralis de ratos submetidos a condições normais de criação desde a eutanásia até 24 horas post-mortem; e (2) determinar se o estresse causado pela contenção dos ratos no período ante-mortem interfere na curva de pH normal da transformação do músculo em carne nos ratos. Para a determinação da curva de pH muscular foram utilizados 12 ratos machos da linhagem Wistar. Os animais sofreram eutanásia e as carcaças foram colocadas a 4°C para as aferições do pH muscular. Outro grupo de 12 animais foi submetido ao estresse por contenção durante uma hora. Após esse período os animais foram eutanasiados e foram realizadas as avaliações de pH. Os resultados indicaram que a curva do pH muscular post-mortem dos ratos apresentaram ritmo de queda semelhante ao descrito para outras espécies comerciais. As médias de pH muscular apresentam diferenças significativas da hora 0 até 6 horas após a eutanásia (p<0,05), quando o valor mínimo de pH muscular foi encontrado. Os ratos em condições de estresse por contenção apresentaram uma queda do pH muscular significativamente mais rápida do que a curva normal (p<0,05).
