Oxidative stress in SH-SY5Y cells on exposure to hydrogen peroxide

Resumo: | Oxidative stress is obtained through the formation of reactive oxygen species, which are normally produced by cell metabolism, but when produced in excess, the cell loses its ability to defend and repair, leading to damage tobiomolecules (DNA, lipids, proteins, etc). When these damages are not repaired, they end up compromising the cell's functioning, leading to death by apoptosis or necrosis. Oxidative stress is involved in neural cell death, which is a major cause of neurodegenerative diseases, including Alzheimer and Parkinson's disease. The aim of this study is to verify the action of hydrogen peroxide as a model of oxidative stress in human neuroblastoma cells. In order to analyze from which concentration a significant stress occurs in the cells and together, verify each concentration at different times of action in the cells. The experiments performed were the analysis of mitochondrial activity obtained through the MTT test, cell viability test using the Trypan blue assay, obtaining quantitative and qualitative data. It is possible to observe that from the concentration of 0.2 mM in the time of 2 hours, there is a significant decrease in the number of cells in relation to the control group, therefore, with the increase of the other concentrations in relation to time, more damage occurs in the cells. |

Cuando estos daños no se reparan, acaban comprometiendo el funcionamiento de la célula, llevándola a la muerte por apoptosis o necrosis. El estrés oxidativo está involucrado en la muerte de las células neurales, que es una de las principales causas de enfermidades neurodegenerativas, incluidas la enfermedad de Alzheimer y la de Parkinson. El objetivo de este estudio es verificar la acción del peróxido de hidrógeno como modelo de estrés oxidativo en células de neuroblastoma humano. Con el fin de analizar a partir de qué concentración se produce un estrés significativo en las células y en conjunto, verificar cada concentración en diferentes tiempos de acción en las células. Los experimentos realizados fueron el análisis de la actividad mitocondrial obtenida a través de la prueba MTT, prueba de viabilidad celular mediante el ensayo de azul de tripano, obteniendo datos cuantitativos y cualitativos. Es posible observar que a partir de la concentración de 0,2 mM en el tiempo de 2 horas, se produce una disminución significativa del número de células con relación al grupo control, por tanto, con el aumento de las demás concentraciones en relación con el tiempo, se produce más daño en las células.O estresse oxidativo é consequência da formação excessiva de espécies reativas de oxigênio geradas normalmente pelo metabolismo celular. Essa produção excessiva ocasiona a perda de capacidade de defesa e de reparo, levando a danos nas biomoléculas (DNA, lipídios, proteínas) celulares. Quando estes danos não são reparados, acabam comprometendo o funcionamento da célula, levando-a a morte por apoptose ou necrose. O estresse oxidativo está associado à morte de células neurais como sendo uma das principais causas de doenças neurodegenerativas, incluindo doença de Alzheimer e doença de Parkinson. O objetivo deste estudo é verificar a ação do peróxido de hidrogênio sendo um modelo de estresse oxidativo em células de neuroblastoma humano. Busca-se analisar a partir de qual concentração ocorre um estresse significante nas células e, em conjunto, verificar esse limiar de concentração em diferentes tempos de ação nas células. Os experimentos realizados compreenderam as análises de atividade mitocondrial obtido por meio do teste de corante tetrazólio (MTT), teste de viabilidade celular utilizando o ensaio de azul de tripano, obtendo-se assim dados quantitativos e qualitativos. Foi possível observar que, a partir da concentração de 0,2 mM durante um intervalo de tempo de 2 horas houve uma diminuição significativa no número de células viáveis em relação ao grupo controle.
