Alteration in redox profile in multiple sclerosis patients

Resumo: Multiple sclerosis is a neurodegenerative, chronic and autoimmune disease that affects the central nervous system,   characterized by the demyelination of neurons preventing nerve impulses from reaching the intended areas. Oxidative stress is an imbalance between the antioxidant defense system and the reactive species present in the body. Both are related in some way that is not yet elucidated. In view of this, the objective was to evaluate the redox profile of patients with multiple sclerosis by making a comparison with healthy individuals. This study was accepted by the Research Ethics Committee of the University of Cruz Alta under the number: 3,531,405. Participated in the study 10 patients with multiple sclerosis, group (EM) and 8 healthy individuals, control group (CT) both groups signed the Free and Informed Consent Form. For analysis, blood samples were collected, which were processed and the plasma was removed. The analyzes were performed according to techniques described in the literature. The results were analyzed using the Mann-Whitney test, considering statistically significant p <0.05. The results found were: increased GSH in the EM group, Catalase without statistical difference between the groups, increased GST in the CT group and increased lipoperoxidation and protein carbonylation in EM. It is concluded that patients with multiple sclerosis have an altered redox profile compared to healthy individuals.

El estrés oxidativo es un desequilibrio entre el sistema de defensa antioxidante y las especies reactivas presentes en el cuerpo. Ambos están relacionados de alguna manera que aún no se ha dilucidado. Ante esto, el objetivo fue evaluar el perfil redox de los pacientes con esclerosis múltiple comparándolo con individuos sanos. Este estudio fue aceptado por el Comité de Ética en Investigación de la Universidad de Cruz Alta bajo el número: 3.531.405. Participaron del estudio 10 pacientes con esclerosis múltiple, grupo (EM) y 8 individuos sanos, grupo control (TC), ambos grupos firmaron el Formulario de Consentimiento Libre e Informado. Para el análisis se recolectaron muestras de sangre, que se procesaron y se extrajo el plasma. Los análisis se realizaron de acuerdo con las técnicas descritas en la literatura. Los resultados se analizaron mediante la prueba de Mann-Whitney, considerando estadísticamente significativa una p <0,05. Los resultados encontrados fueron: aumento de GSH en el grupo EM, catalasa sin diferencia estadística entre los grupos, aumento de GST en el grupo CT y aumento de la lipoperoxidación y carbonilación de proteínas en el grupo EM. Se concluye que los pacientes con esclerosis múltiple tienen un perfil redox alterado en comparación con los individuos sanos.A esclerose múltipla é uma doença neurodegenerativa, crônica e autoimune que atinge o sistema nervoso central, caracterizada pela desmielinização de neurônios impedindo que os impulsos nervosos cheguem até as áreas pretendidas. O estresse oxidativo trata-se de um desequílibrio entre o sistema de defesa antioxidante e as espécies reativas presentes no organismo. Ambos encontram-se relacionados de alguma maneira que todavia não é elucidada. Visto isto, objetivou-se avaliar o perfil redox de portadores de esclerose múltipla realizando uma comparação com indivíduos saudáveis. Este estudo foi aceito pelo Comitê de Ética em Pesquisa da Universidade de Cruz Alta sob parecer de nº: 3.531.405. Participaram do estudo 10 portadores de esclerose múltipla, grupo (EM) e 8 indivíduos saudáveis, grupo controle (CT) ambos os grupos assinaram o Termo de Consentimento Livre e Esclarecido. Para análises foram coletadas amostras sanguíneas, que foram processadas e foi retirado o plasma. As análises foram realizadas conforme técnicas descritas na literatura. Os resultados foram análisados por teste de Mann-whitney considerando estatisticamente significativo p<0,05. Os resultados encontrados foram: GSH aumentada no grupo EM, Catalase sem diferença estatística entre os grupos, GST aumentada no grupo CT e lipoperoxidação e carbonilação proteíca aumentadas em EM.
