Bacteriotech construction in a higher education institution for teaching and research purposes

Resumo: Introduction: Bacteriotecs have one for great relevance to Higher Education Institutions, both for didactic use and for use in research. The storage methods aim to preserve the morphophysiological aspects of the specimens for use at any time. Microbial conservation techniques can be short term, medium term and long term. Among the principles of each technique are cryopreservation methods with common freezing and deep-freezing associated with cryoprotective additives to reduce and protect freeze-related damage. Over the years, the construction of the bacteotecs has been consolidated by providing bacterial bioavailability for educational and research purposes. Objective: This research aimed at building the bacteriotech for the Microbiology Laboratory of the Higher Education Unit of Feira de Santana. Methodology: 14 bacteria were introduced during the research, applying the storage by freezing at -20° C with 20% glycerol and 15% skimmed milk in TSB broth, in addition to the management for maintenance, feasibility and quality control performed in parallel. Results: The technique adopted for cryopreservation in the medium term with the use of two cryoprotectors, as well as the procedures adopted for viability, were able to guarantee for 1 year and 4 months the purity of the available strains without loss of viability. Conclusion: It was noted that the insertion of the bacteriotech was able to meet the academic and scientific needs of the Institution, so the bacteriotech can be inserted with easy and low-cost techniques.

Los métodos de almacenamiento tienen por objeto preservar los aspectos morfofisiológicos de los especímenes para su uso en cualquier momento. Las técnicas de conservación microbiana pueden ser de corto, medio y largo plazo. Entre los principios de cada técnica figuran los métodos de criopreservación con congelación y ultracongelación comunes asociados a aditivos crioprotectores para reducir y proteger los daños relacionados con la congelación. A lo largo de los años, la construcción de la bacteriotech se ha consolidado proporcionando biodisponibilidad bacteriana con fines educativos y de investigación. Objetivo: Esta investigación tenía como objetivo construir la bacterioteca para el Laboratorio de Microbiología de la Unidad de Enseñanza Superior de Feira de Santana. Metodología: Durante la investigación se introdujeron 14 bacterias, aplicando el almacenamiento por congelación a -20° C con 20% de glicerol y 15% de leche desnatada en caldo TSB, además de la gestión para el mantenimiento, la viabilidad y el control de calidad realizados en paralelo. Resultados: La técnica adoptada para la criopreservación a medio plazo con el uso de dos crioprotectores, así como los procedimientos adoptados para la viabilidad, pudieron garantizar durante 1 año y 4 meses la pureza de las cepas disponibles sin pérdida de viabilidad. Conclusión: Se observó que la inserción del bacteriotecnia podía satisfacer las necesidades académicas y científicas de la Institución, por lo que el bacteriotecnia puede insertarse con técnicas fáciles y de bajo costo.Introdução: As bacteriotecas possuem um para grande relevância para Instituições de Ensino Superior, tanto para uso didático, quanto para uso em pesquisas. Os métodos de estocagem visam preservar os aspectos morfofisiológicos dos espécimes para sua utilização a qualquer momento. As técnicas de para conservação microbiana podem ser a curto prazo, médio prazo e longo prazo. Dentre os princípios de cada técnica, os métodos de criopreservação com congelamentos comum e ultracongelamento associados a aditivos crioprotetores para redução e proteção dos danos relacionado ao congelamento. Ao longo dos anos a construção das bacteiotecas vem sendo consolidadas por proporcionar a biodisponibilidade bacteriana para os fins didáticos e de pesquisas. Objetivo: A presente pesquisa objetivou a construção da bacterioteca para o laboratório de Microbiologia da Unidade de Ensino Superior de Feira de Santana. Metodologia: Foram introduzidas no decorrer da pesquisa 14 bactérias, aplicando a estocagem pelo congelamento a -20° C com glicerol a 20% e leite desnatado a 15% em caldo TSB, além da gestão para manutenção, viabilidade e controle de qualidade executadas em paralelo. Resultados: a técnica adotada para a criopreservação em médio prazo com o uso de dois crioprotetores, bem como os procedimentos adotados para viabilidade, poderam garantir durante 1 ano e 4 meses  a pureza das cepas disponíveis sem perda da viabilidade.
