Histochemistry study of the jejunum and the cecum in commercial laying hens submitted to the different forced molting programs

Resumo: | The objective of this study was to assess the alterations in the glycoproteins of the jejuno and the ceco of commercial poetry by histochemical techniques, and they will be subjected to different programs of forced molting. The study occurred in aviary of FCAV-Unesp, Jaboticabal campus. Thirty-two 58-week old Hisex Brown laying hens were used, housed in laying cages (two birds/cage) and randomly distributed in four forced molting programs with two production cycles, ending in 28 and 140 days, respectively. Four birds were allocated to each group, distributed into: Group 1 (G1), in which the California method was used as a control; group 2 (G2), in which a low-calcium diet was used; group 3 (G3), in which a diet with a high level of zinc was provided, and for group 4 (G4), a diet with a low level of sodium. The fragments of jejunum and cecum were submitted to the histochemistry techniques: PAS, PAS + amilase, AB 0,5 and AB 2,5 and were observed the glycoprotein, glycogen, sulfated and carboxyled polysacharides, respectively. The results suggest that the different treatments of forced molting in the experimental conditions did not interfere in the integrity of intestinal cells of these segments. It is concluded that the four treatments of forced molting did not interfere with the presence of intestinal cellular polysaccharides as well as with the integrity of the morphology of the intestinal cells and the glycocalyx. |

El estudio se produjo en el aviario de la FCAV-Unesp, campus de Jaboticabal. Se utilizaron 32 gallinas ponedoras Hisex Brown de 58 semanas de edad, alojadas en jaulas de postura (dos aves / jaula) y distribuidas aleatoriamente en cuatro programas de muda forzada con dos ciclos de producción, finalizando en 28 y 140 días, respectivamente. Se asignaron cuatro aves a cada grupo, distribuidas en: Grupo 1 (G1), en el que se utilizó el método de California como control; grupo 2 (G2), en el que se utilizó una dieta baja en calcio; el grupo 3 (G3), en el que se proporcionó una dieta con alto nivel de zinc, y para el grupo 4 (G4), una dieta con bajo nivel de sodio. Los fragmentos de yeyuno y ciego se sometieron a las técnicas de histoquímica: PAS, PAS + amilasa, AB 0,5 y AB 2,5 y se observaron la glucoproteína, glucógeno, polisacáridos sulfatados y carboxilados, respectivamente. Los resultados sugieren que los diferentes tratamientos de muda forzada en las condiciones experimentales no interfirieron en la integridad de las células intestinales de estos segmentos. Se concluye que los cuatro tratamientos de muda forzada no interfirieron con la presencia de polisacáridos celulares intestinales así como con la integridad de la morfología de las células intestinales y el glucocáliz.O objetivo desse estudo foi avaliar as alterações nas glicoproteínas do jejuno e do ceco de galinhas poedeiras comerciais por técnicas histoquímicas após serem submetidas a diferentes programas de muda forçada. O estudo ocorreu no aviário da FCAV-Unesp, campus Jaboticabal. Foram utilizadas 32 galinhas poedeiras da linhagem Hisex Brown com 58 semanas de vida alojadas em gaiolas de postura (duas aves/gaiola) e distribuídas aleatoriamente em quatro programas de muda forçada com dois ciclos de produção, com término em 28 e 140 dias respectivamente. Para cada grupo foram alocadas quatro aves, distribuídas em: Grupo 1 (G1), no qual utilizou-se o método Califórnia como controle; grupo 2 (G2), no qual utilizou-se dieta com baixo nível de cálcio; grupo 3 (G3), no qual forneceu-se ração com alto nível de zinco e para o grupo 4 (G4), com dieta com baixo nível de sódio. Os fragmentos de jejuno e ceco foram corados mediante as técnicas histoquímicas de PAS, PAS + amilase, AB 0,5 e AB 2,5 no qual observou-se a presença de glicoproteínas, glicogênio, polissacarídeos ácidos sulfatados e carboxilados, respectivamente. Os resultados sugerem que os diferentes tratamentos de muda forçada nas condições experimentais não interferiram na integridade das células intestinais dos dois segmentos intestinais analisados.
