Preparation and characterization of poly(butylene adipate-co-terephthalate) films added with Melaleuca alternifolia essential oil

Resumo: With the aim of prolonging the shelf life and preserving the properties of food, active packaging emerged. In this study, poly(butylene adipate-co-terephthalate) films added with 5, 10 and 15 % (m/m) of Melaleuca alternifolia essential oil were prepared by the solution casting technique. The oil was characterized by hyphenated gas chromatography coupled with mass spectrometry (GC-MS) and Fourier transform mid-infrared spectroscopy (FTIR), identifying as major components: aromadendrene, α-guayene, α-terpineol and 2-methylisoborneol. Oil’s antimicrobial activity was evaluated against the bacteria Escherichia coli by disk-diffusion technique in agar medium, obtaining a halo of 15 mm. The mid-infrared spectra of the films, when subjected to a principal component analysis, showed that there was incoporation of oil into the polymer matrix. The increase in the oil concentration increased the water vapor permeability and reduced the film stiffness, evaluated by mechanical tensile tests. It was also found, through thermogravimetry and differential exploratory calorimetry techniques that the incorporated oil did not change: the thermal stability of the polymer (keeping the degradation in a single step), the melting and crystallization temperatures, the degree of crystallinity of the polymer and the enthalpy of fusion. Finally, the film added with 15% (w/w) of oil showed good inhibition against the Escherichia coli bacteria, after 12 days of contact with a sample of mozzarella cheese at 4º C, reducing the inoculated microbial load of 1,08 ∙ 107 CFU/mL to 2,75 ∙ 106 CFU/mL, which makes this material a promising antimicrobial active packaging.

En este estudio, films de poli (butileno adipato co-tereftalato) aditivados con 5, 10 e 15% de aceite esencial de Melaleuca alternifólia fueron preparados con la técnica de solution casting. El aceite fue caracterizado por cromatografía gaseosa hifenada con espectrometría de masas y espectroscopia en el infrarrojo mediano por transformada de Fourier, identificándose como componentes mayoritarios: aromadendreno, α-guaieno, α-terpineol e 2-metilisoborneol. La actividad antimicrobiana del aceite fue evaluada frente a la bacteria Escherichia coli por la técnica de disco-difusión en agar, obteniendo un halo de 15 mm. Los espectros de infrarrojo mediano de los films, cuándo sometidos al análisis de componentes principales, demostraron que hubo incorporación del aceite en la matriz polimérica. El aumento de la concentración del aceite aumentó la permeabilidad al vapor de agua y redujo la rigidez del film, evaluada por ensayos mecánicos de tracción. También se constató, a través de la termogravimetría y de la calorimetría exploratoria diferencial, que el aceite incorporado no alteró: la estabilidad térmica del polímero (manteniendo la degradación en una única etapa), las temperaturas de fusión y de cristalización, el grado de cristalinidad y la entalpía de fusión. Por fin, el film aditivado con 15% (m/m) del aceite presentó buena inhibición frente a la bacteria Escherichia coli, pasados 12 días de contacto con una muestra de queso muzarella en la temperatura de 4 °C, reduciendo la carga microbiana inoculada de 1,08 ∙ 107 UFC/mL para 2,75 ∙ 106 UFC/mL, lo que hace de este material una promisoria embalaje activa antimicrobiana.Com o intuito de prolongar o tempo de prateleira e preservar as propriedades dos alimentos, surgiram as embalagens ativas. Neste estudo, filmes de poli(butileno adipato-co-tereftalato) aditivados com 5, 10 e 15 % (m/m) de óleo essencial de Melaleuca alternifolia foram preparados pela técnica de solution casting. O óleo foi caracterizado por cromatografia gasosa hifenada com espectrometria de massas e espectroscopia no infravermelho médio por transformada de Fourier, identificando-se como componentes majoritários: aromadendreno, α-guaieno, α-terpineol e 2-metilisoborneol. A atividade antimicrobiana do óleo foi avaliada frente à bactéria Escherichia coli pela técnica de disco-difusão em ágar, obtendo-se um halo de 15 mm. Os espectros de infravermelho médio dos filmes, quando submetidos à análise de componentes principais, mostraram que houve incorporação do óleo na matriz polimérica. O aumento da concentração do óleo aumentou a permeabilidade ao vapor de água e reduziu a rigidez do filme. Também se constatou, através da termogravimetria e da calorimetria exploratória diferencial, que o óleo incorporado não alterou: a estabilidade térmica do polímero (mantendo a degradação em uma única etapa), as temperaturas de fusão e de cristalização, o grau de cristalinidade e a entalpia de fusão.
