Clinical-pathological characterization and research of COX-2, CEA and CA15.3 in cats with breast cancer

Resumo: Mammary gland neoplasms have a multifactorial origin and are classified as the main type of neoformation that affects cats. The diagnosis is obtained from histopathological analysis, where it is possible to determine the type and histological grade of the breast tumor. More recently, tissue biomarkers have allowed more accurate analyzes of the degree of malignancy of tumors, with an attempt to use serum markers to diagnose micronodules and metastases early. Thus, this study aimed to characterize the clinical profile and evaluate the serum levels of carcinoembryonic antigen (CEA) and carcinogen antigen 15.3 (CA15.3), quantified by ELISA. In addition, the objective was to define the histopathological diagnosis and identify the immunohistochemical profile of cyclooxygenase-2 (COX-2) in breast neoplasms. The study was carried out in 49 cats, 38.78% were not neutered and 59.18% had already been submitted to contraceptive hormones. Malignant neoplasms accounted for 76.92%, with the cribriform, tubulopapillary and tubular types being the most frequent. The mean serum levels of CA15.3 and CEA were 2.4108 ± 0.4748 U/mL and 0.0024 µg/L, respectively. The mammary tumor in cats is highly malignant, and more than one histological type can coexist in cats with multiple nodules. The immunoexpression of COX-2 and showed no significant association with the appearance of breast tumors. The use of tumor markers CEA and CA15.3 was not efficient in cats with mammary lesions.

El diagnóstico se obtiene a partir del análisis histopatológico, donde es posible determinar el tipo y grado histológico del tumor mamario. Más recientemente, los biomarcadores tisulares han permitido análisis más precisos del grado de malignidad de los tumores, con un intento de usar marcadores séricos para diagnosticar micronódulos y metástasis de manera temprana. Así, este estudio tuvo como objetivo caracterizar el cuadro clínico y evaluar los niveles séricos de antígeno carcinoembrionario (CEA) y antígeno carcinógeno 15.3 (CA15.3), cuantificados por ELISA. Además, el objetivo fue definir el diagnóstico histopatológico e identificar el perfil inmunohistoquímico de la ciclooxigenasa-2 (COX-2) en las neoplasias de mama. El estudio se realizó en 49 gatas, el 38,78% no estaban esterilizadas y el 59,18% ya habían sido sometidas a hormonas anticonceptivas. Las neoplasias malignas representaron el 76,92%, siendo las más frecuentes las de tipo cribiforme, tubulopapilar y tubular. Los niveles séricos medios de CA15.3 y CEA fueron 2,4108 ± 0,4748 U/mL y 0,0024 µg/L, respectivamente. El tumor mamario en gatas es altamente maligno, pudiendo coexistir más de un tipo histológico en gatas con múltiples nódulos. La inmunoexpresión de COX-2 ya no mostró asociación significativa con la aparición de tumores de mama. El uso de los marcadores tumorales CEA y CA15.3 no fue eficaz en gatas con lesiones mamarias.As neoplasias da glândula mamária têm origem multifatorial e são classificadas como o principal tipo de neoformação que acomete as gatas. Mais recentemente, os biomarcadores teciduais permitiram análises mais precisas quanto ao grau de malignidade dos tumores, havendo uma tentativa de utilizar marcadores séricos para diagnosticar precocemente micronódulos e metástases. Assim, este estudo teve como objetivo caracterizar o perfil clínico e avaliar os níveis séricos de antígeno carcinoembrionário (CEA) e antígeno cancerígeno 15.3 (CA15.3), quantificados por ELISA. Além disso, objetivou-se definir o diagnóstico histopatológico e identificar o perfil imuno-histoquímico da ciclooxigenase-2 (COX-2) nas neoplasias mamárias. O estudo foi realizaodo em 49 gatas, 38,78% não eram castradas e 59,18% já haviam sido submetidas a hormônios contraceptivos. As neoplasias malignas representaram 76,92%, sendo os tipos cribriforme, tubulopapilar e tubular os mais frequentes. Os níveis séricos médios de CA15,3 e CEA foram 2,4108 ± 0,4748 U/mL e 0,0024 µg/L, respectivamente. O tumor mamário nas gatas possui alta malignidade, podendo coexistir mais de um tipo histológico em gatas com múltiplos nódulos. A imunoexpressão de COX-2 e não mostrou associação significativa com o aparecimento de tumores mamários.
