Antitumor and human topoisomerase II-α inhibitory activity II-α of the flavonoid luteolin isolated from Prosopis juliflora

Resumo: | This study aimed to evaluate the antitumor and inhibitory activity of human topoisomerase II-α of the flavonoid luteolin isolated from Prosopis juliflora against prostate tumor cells (PC-3), with the aim of investigating new drugs of natural origin. In vitro tests were conducted with crude leaf extracts against PC-3 and FGH cell lines. Cytotoxicity tests were performed using MTT (3-(4,5-dimethiazol-2yl) 2,5-diphenyl tetrazolium bromide) with monitoring after 24 h of exposure of tumor and non-tumor cells to different concentrations of extracts (100; 50; 25; 10 μg/mL). Luteolin extracted from the leaves of P. juliflora showed positive results on the DNA topo II-α enzyme essential in the replication process of cancer cells from the 50 µM concentration. Furthermore, it showed antitumor activity against PC-3 cells from the lowest concentration tested 10 µM, and absence of cytotoxicity against normal cells. A chemical study for the extraction and isolation of active compounds from P. juliflora leaf extracts is suggested, to investigate the possible antitumor activity of these compounds in in vitro and in vivo assays. |

Se realizaron pruebas in vitro con extractos de hojas crudas contra líneas celulares PC-3 y FGH. Se realizaron pruebas de citotoxicidad utilizando MTT (bromuro de 3-(4,5-dimetiazol-2il) 2,5-difeniltetrazolio) con seguimiento a las 24 h de exposición de células tumorales y no tumorales a diferentes concentraciones de extractos (100; 50; 25; 10 μg/mL). La luteolina extraída de las hojas de P. juliflora mostró resultados positivos en la enzima ADN topo II-α esencial en el proceso de replicación de las células cancerosas a partir de la concentración de 50 µM. Además, mostró actividad antitumoral frente a células PC-3 desde la concentración más baja ensayada 10 µM, y ausencia de citotoxicidad frente a células normales. Se sugiere un estudio químico para la extracción y aislamiento de compuestos activos de extractos de hojas de P. juliflora, para investigar la posible actividad antitumoral de estos compuestos en ensayos in vitro e in vivo.Este estudo objetivou avaliar a atividade antitumoral e inibitória da topoisomerase II-α humana do flavonoide luteolina isolado de Prosopis juliflora frente a células tumorais prostáticas (PC-3), com o intuito de investigação de novos fármacos de origem natural. Testes in vitro foram conduzidos com os extratos brutos das folhas frente as linhagens de células PC-3 e linhagem FGH. Os testes de citotoxicidade foram realizados através de MTT (3- (4,5-dimetiazol-2il) 2,5-difenil tetrazólio de brometo) com monitorização após 24 h de exposição das células tumorais e não tumorais a diferentes concentrações dos extratos (100; 50; 25; 10 μg/mL). A luteolina extraída das folhas da P. juliflora apresentou resultados positivos sobre a inibição de DNA topo II-α enzima essencial no processo de replicação das células cancerígenas a partir da concetração 50 µM. Ademais, apresentou atividade antitumoral frente a células PC-3 a partir da menor concentração testada 10 µM, e ausência de citotoxicidade frente a células normais.
