Amniotic membrane reduces the alveolar bone loss and increases attachment level gain in periodontal disease in rats

Resumo: | Periodontal disease is a multifactorial chronic inflammatory process that leads to the destruction of periodontium structures and later tooth loss, impairing oral and general health. Amniotic membrane (AM) attracts attention as a biomaterial for regenerative medicine; its unique architecture ensures excellent biological and mechanical properties. The objective is to analyze the potential of AM in the regeneration of periodontal tissues in induced periodontitis in rats. The sample involved 26 male Rattus norvegicus, Albinos, Wistar rats, weighing approximately 200 to 250g, which after the acclimatization period were randomly divided into groups: Normal group (n=6): the animals were not submitted to any procedure , following the normal course of the experiment; Ligation group (n=10): the animals were submitted to ligation for induction of periodontitis, and after 2 weeks it was removed and continued without treatment; Ligation + AM group (n=10): the animals were submitted to ligation to induce periodontitis, after 2 weeks the ligature was removed and the AM placed around the bone defect. After 4 weeks of AM action, the animals were euthanized and macroscopic, morphometric, histological, histomorphometric and statistical analyzes were performed to evaluate periodontal repair. There was a significant installation of periodontal disease, and after the placement of AM, there was a decrease in alveolar bone loss and probing depth, gain in the level of conjunctival attachment, and periodontal tissues with an aspect of normality. It is concluded that AM stimulates periodontal repair after removing the etiologic agent that was causing periodontitis. |

La membrana amniótica (AM) atrae la atención como biomaterial para la medicina regenerativa, su arquitectura única asegura excelentes propiedades biológicas y mecánicas. El objetivo es analizar el potencial de la MA en la regeneración de tejidos periodontales en periodontitis inducida en ratas. La muestra involucró a 26 ratas macho Rattus norvegicus, Albinos, Wistar, con un peso aproximado de 200 a 250 g, que después del período de aclimatación se dividieron aleatoriamente en grupos: Grupo normal (n=6): los animales no fueron sometidos a ningún procedimiento, siguiendo el curso del experimento; Grupo ligadura (n=10): los animales fueron sometidos a ligadura para inducción de periodontitis, ya las 2 semanas se retiró y se continuó sin tratamiento; Grupo Ligadura + MA (n=10): los animales fueron sometidos a ligadura para inducir la periodontitis, después de 2 semanas se retiró la ligadura y se colocó la MA alrededor del defecto óseo. Después de 4 semanas de acción AM, los animales fueron sacrificados y se realizaron análisis macroscópicos, morfométricos, histológicos, histomorfométricos y estadísticos para evaluar la reparación periodontal. Hubo una instalación importante de enfermedad periodontal, y después de la colocación de AM, hubo una disminución en la pérdida de hueso alveolar y profundidad de sondaje, ganancia en el nivel de inserción conjuntival y tejidos periodontales con un aspecto de normalidad. O objetivo é analisar o potencial da MA na regeneração dos tecidos periodontais na periodontite induzida em ratos. A casuística envolveu 26 ratos machos Rattus norvegicus, Albinos, Wistar, com peso aproximado de 200 a 250g, os quais após o período de aclimatização foram divididos aleatoriamente nos grupos: Grupo normal (n=6): os animais não foram submetidos a nenhum procedimento, seguindo o percurso normal do experimento; Grupo ligadura (n=10): os animais foram submetidos à ligadura para indução da periodontite, e após 2 semanas esta foi removida e seguiram sem tratamento; Grupo ligadura + MA (n=10): os animais foram submetidos a ligadura para indução de periodontite, após 2 semanas a ligadura foi removida e a MA colocada ao redor do defeito ósseo. Após 4 semanas de atuação da MA os animais foram eutanasiados e as análises macroscópica, morfométrica, histológica, histomorfométrica e estatística foram realizadas para avaliar o reparo periodontal. Observou-se significativa instalação da doença periodontal, e após a colocação da MA, verificou-se a diminuição da perda óssea alveolar e da profundidade de sondagem, ganho no nível de inserção conjuntiva, e tecidos periodontais com aspecto de normalidade.
