Antifungal potential of extracts of the microalgae Pyramimonas virginica against dermatophyte fungi

Resumo: | The aim of this study was to evaluate the antifungal potential of the extracts centrifuged and electroflocculated of the microalgae Pyramimonas virginica against dermatophyte fungi. To obtain the microalgal biomass, the culture was carried out in a 10 L tubular photobioreactor, in triplicates. The harvesting of microalgal biomass was performed by two methods, centrifugation and electroflocculation. For the extraction of microalgal biomass, the absolute ethanol (99.9%) solvent was used. The antifungal activity of the extracts was evaluated by broth microdilution method, in the concentration range of 0.0115 to 6 mg mL-1. The Minimum Inhibitory Concentration (MIC) was determined visually, by the absence of fungal growth. Extracts were tested against the dermatophytes Nannizzia gypsea, Trichophyton mentagrophytes and Trichophyton tonsurans. Both centrifuged and electroflocculated extracts were able to inhibit the growth of all dermatophytes evaluated, and the centrifuged extract was more active than the electroflocculated. The extracts showed lower MIC values against T. tonsurans (MIC: 0.75 mg mL-1) and N. gypsea (MIC: 0.75 mg mL-1), followed by T. mentagrophytes (MIC: 6 mg mL-1). Regarding the type of activity recorded, against the fungus T. mentagrophytes it was of the fungiostatic type and for the fungi T. tonsurans and N. gypsea it was of the fungicide type. This was a pioneering study on the antifungal activity of the microalgae P. virginica, as well as the method of fractionation of its biomass. The microalgae demonstrated antifungal potential against the dermatophyte fungi evaluated, showing to be promising in the development of new herbal medicines and treatments. |

La producción de la biomasa de microalga fue obtenida a partir del cultivo en fotobiorreactor tubular (10 L). La biomasa de microalgas se recogió por medio de centrifugación y electrofloculación. La actividad antifúngica de los extractos se evaluó por medio del método de microdilución en caldo, en el rango de concentración de 0.0115 a 6 mg mL-1. La Concentración Mínima Inhibitoria (CMI) se definió por la ausencia de crecimiento fúngico. Se han probado los extractos de la microalga frente a los dermatofitos Nannizzia gypsea, Trichophyton mentagrophytes y Trichophyton tonsurans. Los extractos de microalgas obtenidos por centrifugado y electrofloculado, lograron inhibir el crecimiento de todos los dermatofitos evaluados, siendo el extracto centrifugado más activo que el electrofloculado. Los extractos tuvieron el valor de CIM más bajo frente a T. tonsurans (CIM: 0.75 mg mL-1) y N. gypsea (CIM: 0.75 mg mL-1), seguidos de T. mentagrophytes (CIM: 6 mg mL-1). La actividad registrada, frente al hongo T. mentagrophytes fue del tipo fungistática y para los hongos T. tonsurans y N. gypsea fue del tipo fungicida. Se trata de un estudio pionero respecto la actividad antifúngica de la microalga P. virginica, así como sobre el método de fraccionamiento de su biomasa. En este trabajo se evidencia el potencial antifúngico contra los hongos dermatofitos evaluados, que se muestra prometedor en el desarrollo de nuevos medicamentos y tratamientos.O objetivo deste estudo foi avaliar o potencial antifúngico in vitro dos extratos etanólicos, centrifugado e eletrofloculado, da microalga Pyramimonas virginica frente a fungos dermatófitos. Para obtenção da biomassa microalgal, o cultivo foi realizado em fotobiorreator tubular de 10 L, em triplicatas. A coleta da biomassa microalgal foi realizada por dois métodos, centrifugação e eletrofloculação. Para extração da biomassa microalgal, foi utilizado o solvente etanol absoluto (99.9%). A atividade antifúngica dos extratos foi avaliada pelo método de microdiluição em caldo, no intervalo de concentração de 0.0115 a 6 mg mL-1. A Concentração Inibitória Mínima (CIM) foi determinada visualmente, pela ausência de crescimento do fungo. Os extratos foram testados contra os dermatófitos Nannizzia gypsea, Trichophyton mentagrophytes e Trichophyton tonsurans. Ambos os extratos microalgais, centrifugado e eletrofloculado, conseguiram inibir o crescimento de todos os dermatófitos avaliados, sendo o extrato centrifugado mais ativo do que o extrato eletrofloculado. Os extratos apresentaram menor valor de CIM contra T. tonsurans (CIM: 0.75 mg mL-1) e N. gypsea (CIM: 0.75 mg mL-1), seguido por T. mentagrophytes (CIM: 6 mg mL-1). Quanto ao tipo de atividade registrada, frente ao fungo T. mentagrophytes a atividade foi do tipo fungiostática e para os fungos T. tonsurans e N. gypsea foi do tipo fungicida. Este foi um estudo pioneiro acerca da atividade antifúngica da microalga P. virginica, assim como do método de fracionamento da sua biomassa.
