The antimicrobial properties of the glycolic extract of Casearia sylvestris Sw. in monotypic biofilm of Candida albicans and Candida glabrata

Resumo: Objectives: The resistance of microorganisms to the misuse of drugs is currently one of the public health problems, making research on natural compounds with antimicrobial activity, such as Casearia sylvestris, necessary. This study aimed to evaluate the antimicrobial activity of the glycolic extract of Casearia sylvestris on standard strains of Candida albicans (ATCC 18804) and Candida glabrata (ATCC 9030) in planktonic cultures, verifying the minimum inhibitory concentration and minimum microbicide concentration (MIC and CMM) and also its action on biofilms. Methodology: The glycolic extract of C. sylvestris, of commercial origin, was obtained at a concentration of 200 mg/ml (20%) eluted in 80% of propylene glycol. The MIC and CMM values were determined by the broth microdilution method, according to the Clinical and Laboratory Standards Institute (CLSI), standards M27-A2, with modifications. Then, tests were started on 48h monotypic biofilms, with broth change every 24h, of these microorganisms. After undergoing treatments, the biofilm was prepared for the MTT metabolic test. Results: Candida albicans presented MMC at 50 mg/mL, in Candida glabrata there was no MIC and MMC. In the MTT test the results showed effectiveness of the concentration of 25 mg/mL on Candida glabrata, with a reduction of ≥ 29%, in Candida albicans there were no significant results in the analyzed concentrations. Conclusion: The extract hawed an antimicrobial effect against the tested genre. We also emphasize the importance of in vitro and in vivo tests that corroborate the results presented, in order to increase the basis for the application of the extract.

Este estudio tuvo como objetivo evaluar la actividad antimicrobiana del extracto glicólico de Casearia sylvestris sobre cepas estándar de Candida albicans (ATCC 18804) y Candida glabrata (ATCC 9030) en cultivos planctónicos, verificando la concentración mínima inhibitoria y la concentración mínima de microbicida (MIC y CMM) y también su acción sobre biopelículas. Metodología: El extracto glicólico de C. sylvestris, de origen comercial, se obtuvo a una concentración de 200 mg / ml (20%) eluido en 80% de propilenglicol. Los valores de MIC y CMM se determinaron por el método de microdilución en caldo, según el Clinical and Laboratory Standards Institute (CLSI), estándares M27-A2, con modificaciones. A continuación, se iniciaron las pruebas en biofilms monotípicos de 48 h, con cambio de caldo cada 24 h, de estos microorganismos. Después de someterse a tratamientos, la biopelícula se preparó para la prueba metabólica MTT. Resultados: Candida albicans presentó MMC a 50 mg / mL, en Candida glabrata no hubo MIC y MMC. En la prueba MTT los resultados mostraron efectividad de la concentración de 25 mg / mL en Candida glabrata, con una reducción de ≥ 29%, en Candida albicans no hubo resultados significativos en las concentraciones analizadas. Conclusión: El extracto tuvo un efecto antimicrobiano contra el género probado. Este estudo objetivou avaliar a atividade antimicrobiana do extrato glicólico de Casearia sylvestris sobre cepas-padrão de Candida albicans (ATCC 18804) e Candida glabrata (ATCC 9030) em cultura planctônica, verificando a concentração inibitória mínima e concentração microbicida mínima (CIM e CMM) e também sua ação sobre biofilmes. Metodologia: O extrato glicólico de C. sylvestris, de origem comercial, foi obtido na concentração de 200 mg/ml (20%) eluídos em 80% de propilenoglicol. Os valores da CIM e CMM foram determinados pelo método de microdiluição em caldo, segundo Clinical and Laboratory Standards Institute (CLSI), normas M27-A2, com modificações. Logo, foram iniciados os testes em biofilmes monotípicos de 48h, com troca de caldo a cada 24h, destes micro-organismos. Após submetido aos tratamentos o biofilme foi preparado para o teste metabólico de MTT. Resultados: Candida albicans apresentou CMM em 50 mg/mL, em Candida glabrata não observou-se CIM e CMM. No teste de MTT os resultados mostraram efetividade da concentração de 25 mg/mL sobre Candida glabrata, com redução de ≥ 29%, em Candida albicans não observou-se resultados significativos nas concentrações analisadas. Conclusão: O extrato apresentou efeito antimicrobiano contra os gêneros testados.
