Evaluation of the efficacy of biocides in removing biofilms produced by multidrug-resistant Pseudomonas aeruginosa

Resumo: Pseudomonas aeruginosa is one of the most frequent pathogens in infections affecting burn patients, having as an important determinant of pathogenicity its ability to form biofilms. The present work aims to evaluate the effectiveness of biocidal solutions: 4% chlorhexidine, 1% sodium hypochlorite, 5% hydrogen peroxide and 70% alcohol on removal P. aeruginosa multidrug-resistant biofilms. This is a descriptive study with a quantitative approach. Biofilms from nine strains of P. aeruginosa, of different clonal types, were evaluated. Eight strains were carbapenem-resistant and one susceptible, collected from burned patients and balneotherapy tables. Strains were grown together with stainless steel coupons and incubated for 24 h. Subsequently, the coupons were treated with and without biocides, at contact times of 15, 5 and 0 minutes. Biofilm removal was assessed by counting viable cells obtained from coupons with and without treatment. The minimum inhibitory concentration for imipenem and meropenem was determined by using the epsilometric method. Biofilm formation was observed in all coupons not treated with biocides. However, there was no count of viable cells after contact of the biofilm with all biocidal solutions, in all times. The minimum inhibitory concentration to carbapenems varied between 16 to ≥ 32µg / mL. It was concluded that all the disinfectant solutions evaluated were effective in removing biofilms produced by the studied P. aeruginosa strains.

El presente trabajo tiene como objetivo evaluar la efectividad de las soluciones biocidas: clorhexidina al 4%, hipoclorito de sodio al 1%, peróxido de hidrógeno al 5% y alcohol al 70% en la eliminación de biopelículas de P. aeruginosa multidrogas resistentes. Este es un estudio descriptivo con un enfoque cuantitativo. Se evaluaron biopelículas de nueve cepas de P. aeruginosa, de diferentes tipos clonales, ocho resistentes a carbapenems y una sensible, recolectada de pacientes quemados y tablas de balneoterapia. Las cepas estudiadas se cultivaron junto con cupones de acero inoxidable, y se incubaron durante 24 h. Posteriormente, los cupones se trataron con y sin biocidas, en tiempos de contacto de 15, 5 y 0 minutos. La eliminación de la biopelícula se evaluó contando las células viables obtenidas de los cupones con y sin tratamiento. La concentración inhibitoria mínima para imipenem y meropenem se determinó para cepas evaluadas usando el método epsilométrico. Se observó la formación de biopelículas en todos los cupones no tratados con biocidas. Sin embargo, no hubo recuento de células viables después del contacto de la biopelícula con todas las soluciones biocidas, en los tiempos estudiados. La concentración inhibitoria mínima para los carbapenémicos varió entre 16 y ≥ 32 µg / ml. Se concluyó que todas las soluciones desinfectantes evaluadas fueron efectivas para eliminar las biopelículas producidas por las cepas estudiadas de P. aeruginosa.Pseudomonas aeruginosa é um dos patógenos mais frequentes em infecções acometendo pacientes queimados, tendo como importante determinante de patogenicidade sua capacidade de formar biofilmes. A remoção do biofilme foi avaliada através da contagem de células viáveis obtidas dos cupons com e sem tratamento.  A concentração mínima inibitória ao imipenem e meropenem foi determinada para cepas avaliadas através do método epsilométrico. Foi observado formação de biofilme em todos os cupons não tratados com biocidas. Entretanto, não houve contagem de células viáveis após contato do biofilme com todas soluções biocidas, nos tempos estudados. A concentração mínima inibitória aos carbapenêmicos variou entre 16 a ≥ 32µg/mL.
