Infection by bovine Alphaherpesvirus 1 and 5 and the induction of mucosal immunity

Resumo: | The immune system present in the mucosa represents the initial barrier against infection by several pathogens that use these surfaces as a gateway to the host organism, such as the case of bovine Alphaherpesvirus 1 (BoHV-1) and bovine Alphaherpesvirus 5 (BoHV-5). These viruses infect the reproductive and respiratory systems of cattle and are responsible for cases of Infectious Bovine Rhinotracheitis, Infectious Pustular Vulvovaginitis and Infectious Pustular Balanopostitis, as well as Bovine Herpetic Meningoencephalitis, respectively. Infection by BoHVs occurs in cavities lined with mucosal epithelial cells, mainly nasal and genital, being the initial point of replication, followed by local dissemination, eventual viremia and neuronal dissemination, until latency is established. The cooperation of defense mechanisms provides protection through the integrated mucosal system through the activation of effector sites and especially the production of different isotypes of immunoglobulins, such as SIgA, which neutralizes mucosal surfaces, and IgG, which plays a neutralization in the mucosa and systemically. The induction of immunity on mucosal surfaces through the use of vaccines is extremely important for the prevention of virus adsorption to the cell and infection by pathogenic microorganisms and depends, especially, on the immunogen, immunization route and adjuvant substance used in the formulations. This review aimed to address the main aspects related to infection by BoHV-1 and BoHV-5, as well as the types of response related to the mucosal immune system against these viruses, and also studies related to the use of the mucosal pathways for administration of vaccines in different animal species. |

Estos virus infectan los sistemas reproductivo y respiratorio del ganado y son responsables de casos de rinotraqueítis infecciosa bovina, vulvovaginitis pustulosa infecciosa y balanopostitis pustulosa infecciosa, así como meningoencefalitis herpética bovina, respectivamente. La infección por BoHVs ocurre en cavidades revestidas por células epiteliales mucosas, principalmente nasales y genitales, siendo el punto de partida de la replicación, seguida de diseminación local, eventual viremia y diseminación neuronal, hasta que se establece la latencia. La cooperación de los mecanismos de defensa brinda protección a través del sistema mucoso integrado a través de la activación de sitios efectores y especialmente la producción de diferentes isotipos de inmunoglobulinas, como SIgA, que neutraliza las superficies mucosas, e IgG, que juega neutralización en la mucosa y sistémicamente. La inducción de inmunidad en las superficies mucosas mediante el uso de vacunas es extremadamente importante para la prevención de la adsorción de virus a la célula y la infección por microorganismos patógenos y depende, especialmente, del inmunógeno, vía de inmunización y sustancia adyuvante utilizada en las formulaciones. Esta revisión tuvo como objetivo abordar los principales aspectos relacionados con la infección por BoHV-1 y BoHV-5, así como los tipos de respuesta relacionados con el sistema inmunológico de la mucosa frente a estos virus, y también los estudios relacionados con el uso de las vías mucosas de administración. de vacunas en diferentes especies animales.O sistema imune presente nas mucosas representa a barreira inicial frente à infecção por diversos patógenos que utilizam estas superfícies como porta de entrada no organismo hospedeiro, como é o caso dos Alphaherpesvirus bovino 1 (BoHV-1) e Alphaherpesvirus bovino 5 (BoHV-5). A infecção pelos BoHVs ocorre nas cavidades revestidas por células epiteliais de mucosa, principalmente nasal e genital, sendo o ponto inicial de replicação, seguindo-se a disseminação local, viremia eventual e disseminação neuronal, até o estabelecimento da latência. A cooperação dos mecanismos de defesa confere proteção através do sistema integrado de mucosas por meio da ativação dos sítios efetores e especialmente produção de diferentes isotipos de imunoglobulinas, como a SIgA, realiza neutralização nas superfícies mucosas, e a IgG, que desempenha neutralização na mucosa e sistemicamente. A indução da imunidade nas superfícies mucosas através do uso de vacinas torna-se de extrema importância para a prevenção da adsorção do vírus à célula e infecção pelos microrganismos patogênicos e depende, especialmente, do imunógeno, via de imunização e substância adjuvante empregada nas formulações.
