Bioprospection of bromelain extract plus ACP® on buck semen quality

Resumo: | The aim of this study was to evaluate the effect of bromelain extract on sperm quality after thawing in goats. For that, five Anglo Nubian goats, with reproductive age, clinically healthy, were used. Semen collections were performed. After concentration analysis, the total pool volume was divided into five groups. One belonging to the control group, composed of (ACP-101/102®) and four experimental groups with ACP-101/102® enriched with bromelain extract at 5% concentrations; 10%; 15% and 20%. The samples were cryopreserved with the aid of the Tk3000® device. After a period of seven days, the samples were thawed and submitted to evaluations by the CASA system. thermoresistance, fluorescent probes, comet test and ultrastructural sperm analysis test using transmission electron microscopy (TEM). In the evaluation of post-thaw sperm kinetics, it was possible to observe that the parameters of total sperm motility, the 5% bromelain group, stood out among the others, whereas in the curvilinear velocity and mean trajectory velocity the control group presented the best results. As for the integrity of sperm DNA, the concentrations of bromelain (5%, 10%, 15% and 20%) showed no significant difference in relation to the control group, demonstrating that this substance does not present genotoxicity to sperm cells. In the ultrastructural analysis, the 5% bromelain group showed the best results. In this sense, bromelain extract added to diluent media, aiming at cryopreservation, is characterized as a promising substance, especially in maintaining the integrity of sperm DNA. However, more studies are needed for its standardization. |

Para ello se utilizaron cinco cabras anglo nubias, en edad reproductiva, clínicamente sanas. Se realizaron recolecciones de semen Después del análisis de concentración, el volumen total de la piscina se dividió en cinco grupos. Uno perteneciente al grupo control, compuesto por (ACP-101 / 102®) y cuatro grupos experimentales con ACP-101 / 102® enriquecido con extracto de bromelina al 5% de concentración; 10%; 15% y 20%. Las muestras fueron criopreservadas con la ayuda del dispositivo Tk3000®, luego de un período de siete días, las muestras fueron descongeladas y sometidas a evaluación por el sistema CASA. termorresistencia, sondas fluorescentes, prueba de cometa y prueba de análisis ultraestructural de espermatozoides mediante microscopía electrónica de transmisión (TEM). En la evaluación de la cinética espermática post-descongelación, se pudo observar que los parámetros de motilidad espermática total, el grupo bromelina al 5%, sobresalieron entre los demás, mientras que en la velocidad curvilínea y la velocidad media de trayectoria el grupo control presentó la Mejores resultados. En cuanto a la integridad del ADN espermático, las concentraciones de bromelina (5%, 10%, 15% y 20%) no mostraron diferencia significativa en relación al grupo control, demostrando que esta sustancia no presenta genotoxicidad a los espermatozoides. En el análisis ultraestructural, el grupo de bromelina al 5% mostró los mejores resultados. En este sentido, el extracto de bromelina añadido a los medios diluyentes, con el objetivo de la criopreservación, se caracteriza por ser una sustancia prometedora, especialmente para mantener la integridad del ADN espermático. Sin embargo, se necesitan más estudios para su estandarización.Objetivou-se avaliar o efeito do extrato de bromelina sobre a qualidade espermática, pós-descongelação na espécie caprina. Para tanto, foram utilizados cinco caprinos da raça Anglo Nubiana, com idade reprodutiva, clinicamente saudáveis. Foram realizadas colheitas de sêmen. Após a análise da concentração, o volume total do pool foi dividido em cinco grupos. Um pertencente ao grupo controle, composto de (ACP-101/102®) e quatro grupos experimentais com ACP-101/102® enriquecidos com extrato de bromelina nas concentrações de 5%; 10%; 15% e 20%. As amostras foram criopreservadas com auxílio do aparelho Tk3000®. Após o período de sete dias as amostras foram descongeladas e submetidas a avaliações pelo sistema CASA. Termo resistência, sondas fluorescente, teste cometa e teste de análise ultraestrutural de espermatozoides por meio de microscopia eletrônica de transmissão (MET). Na avaliação da cinética espermática pós-descongelação foi possível observar que os parâmetros motilidade espermática total, o grupo bromelina a 5%, se destacou dentre os demais, já na velocidade curvilínea e velocidade média da trajetória o grupo controle apresentou os melhores resultados. Quanto à integridade do DNA espermático as concentrações de bromelina (5%; 10%; 15% e 20%) não apresentaram diferença significativa em relação ao grupo controle, demonstrando que essa substância não apresenta genotoxicidade às células espermáticas. Na análise ultraestrutural o grupo bromelina a 5% demonstrou os melhores resultados. Nesse sentido, o extrato de bromelina adicionada a meios diluentes, visando à criopreservação, caracteriza-se como uma substância promissora, sobretudo da manutenção da integridade do DNA espermático.
