Evaluation of the antibacterial activity of a dentin biomodifier based on Copaifera multijulga Hayne oil

Resumo: Objectives: To evaluate in vitro the antibacterial activity of a dentin biomodifier based on copaiba oil. Methodology: The antibacterial activity test was carried out over a period of 12 months, where the emulsion was stored in different environments and analysed at 0, 3 and 12 months. For that, the microorganisms Streptococcus mutans and Lactobacillus paracasei were used. The positive control was 2% chlorhexidine digluconate and the negative control was the emulsion vehicles: distilled water, 0.5% diazolidinyl urea and 1% polysorbate 80. 96-well microplates were used, so that each well had a final volume of 100 µL. On the microplate, the columns were distributed in the numbers from 1 to 12 and the lines in the letters “A” to “G”. In the wells, the emulsion was distributed at different concentrations, in addition to the negative control and positive control. Results: The emulsion showed bactericidal or bacteriostatic activity against all microorganisms tested at all concentrations for the microorganisms S. mutans and L. paracasei at time zero. After 12 months, the emulsion formulation based on copaiba oil continued to exert bactericidal and bacteriostatic activity at concentrations of 26.66 μL/mL and 15 μL/mL, respectively, against the microorganism S. mutans. The refrigerator environment showed bacteriostatic activity, while the air conditioning environment showed bactericidal activity against S. mutans in the period of 12 months. Conclusion: The biomodifier showed bactericidal and/or bacteriostatic activity in different environments for 12 months, and can be stored in a refrigerator.

Metodología: La prueba de actividad antibacteriana se realizó durante 12 meses, donde la emulsión se almacenó en diferentes ambientes y se analizó a los 0, 3 y 12 meses. Para ello se utilizaron los microorganismos Streptococcus mutans y Lactobacillus paracasei. El control positivo fue digluconato de clorhexidina al 2% y el control negativo fueron los vehículos de la emulsión: agua destilada, diazolidinilurea al 0,5% y polisorbato 80 al 1%. Se utilizaron microplacas de 96 pocillos, de forma que cada pocillo tuviera un volumen final de 100 µL. En la microplaca, las columnas se distribuyeron en los números del 1 al 12 y las líneas en las letras “A” a la “G”. En los pocillos se distribuyó la emulsión a diferentes concentraciones, además del control negativo y el control positivo. Resultados: La emulsión mostró actividad bactericida o bacteriostática contra todos los microorganismos probados a todas las concentraciones para los microorganismos S. mutans y L. paracasei en el tiempo cero. Después de 12 meses, la formulación de la emulsión a base de aceite de copaiba continuó ejerciendo actividad bactericida y bacteriostática a concentraciones de 26,66 μL/mL y 15 μL/mL, respectivamente, contra el microorganismo S. mutans. El ambiente de refrigerador mostró actividad bacteriostática, mientras que el ambiente de aire acondicionado mostró actividad bactericida contra S. mutans en el periodo de 12 meses. Conclusión: El biomodificador mostró actividad bactericida y/o bacteriostática en diferentes ambientes durante 12 meses, pudiendo almacenarse en refrigeración.Objetivos: Avaliar in vitro a atividade antibacteriana de um biomodificador dentinário à base do óleo de copaíba. Metodologia: O teste de atividade antibacteriana foi realizado no período de 12 meses, onde a emulsão foi armazenada em diferentes ambientes e analisada em 0, 3 e 12 meses. Para tanto, foram utilizados os micro-organismos Streptococcus mutans e Lactobacillus paracasei. O controle positivo foi o digluconato de clorexidina a 2% e o controle negativo os veículos da emulsão: água destilada, diazolidinil uréa a 0,5% e polisorbato 80 a 1%. Foram utilizadas microplacas de 96 orifícios, de forma que cada poço possuísse um volume final de 100 µL. Na microplaca, as colunas foram distribuídas nos números de 1 a 12 e as linhas nas letras “A” até “G”. Nos poços, a emulsão foi distribuída nas diferentes concentrações, além do controle negativo e do controle positivo. Resultados: A emulsão apresentou atividade bactericida ou bacteriostática frente a todos os microrganismos testados em todas as concentrações para os microrganismos S. mutans e L. paracasei no tempo zero. Após 12 meses, a formulação da emulsão à base do óleo de copaíba permaneceu exercendo atividade bactericida e bacteriostática nas concentrações 26,66 μL/mL e 15 μL/mL, respectivamente, frente ao microrganismo S. mutans. O ambiente geladeira apresentou-se com atividade bacteriostática, enquanto o ambiente ar condicionado mostrou-se com atividade bactericida frente ao S. mutans no período de 12 meses.
