Physico-chemical characteristics and effect on oxidative stress of red pitaya in swiss diabetic mice

Resumo: Red pitaya (Hylocereus polyrhizus Weber Britton & Rose) is rich in antioxidants, which might help regulate oxidative stress, exacerbated in Diabetes Mellitus (DM). This study aimed to determine the centesimal composition, physicochemical characteristics and in vivo antioxidant effect of red pitaya in diabetic swiss mice. Pulp, seeds and peel were isolated and used for physicochemical and centesimal composition analysis. DM was induced in female Swiss mice through intraperitoneal Alloxan Monohydrate injection. Groups with 6 animals were formed: Healthy (SAUD), Untreated A and B (DNT A and DNT B), Metformin (MET), and groups fed with 20 (PIT20), 40 (PIT40), 200 (PIT200) and 400 (PIT400) mg of pulp with seeds extract/kg of animals’ body weight, by gavage, for 28 days. Thiobarbituric acid reactive species (TBARS) hepatic content was quantified by Malondialdehyde (MDA), to evaluate lipid peroxidation. Pitaya’s composition was similar to that previously reported. Pulp and peel showed insignificant lipid contents, expressive presence of carbohydrates, and fibers in all samples, especially peel. Lowest MDA was observed in PIT400, PIT200, and PIT40, compared to DNT A and B, and MET. Results were dose-dependent, suggesting pitaya consumption was associated with lower lipid peroxidation. Further studies on its composition and applications as a functional food are required, especially for humans and in DM.

Este trabajo tuvo como objetivo la análisis la composición centesimal, de las características fisicoquímicas y del efecto antioxidante in vivo dela pitaya roja en ratones diabeticos. La pulpa, las semillas y la cáscara fueron aisladas y usadas en el análisis de composición fisicoquímica y centesimal. Se indujo DM en ratones hembra suizos mediante inyección intraperitoneal de monohidrato de alloxano. Se formaron grupos con 6 animales: sanos (SAUD), no tratados A y B (DNT A y DNT B), metformina (MET), y grupos alimentados con 20 (PIT20), 40 (PIT40), 200 (PIT200) y 400 (PIT400) mg del extracto de pulpa y semillas/kg de peso corporal de los animales, por gavage, durante 28 días. El contenido hepático de especies reactivas al ácido tiobarbitúrico (TBARS) se cuantificó por malondialdehído (MDA), para evaluar la peroxidación lipídica. La composición de pitaya fue similar a la anteriormente reportada. La pulpa y la cáscara mostraron contenido lipídico insignificante y presencia expresiva de carbohidratos y fibras en todas las muestras, especialmente la cáscara. El MDA más bajo se observó en PIT400, PIT200 y PIT40, en comparación con DNT A y B, y MET. Los resultados fueron dosis-dependientes, lo que sugiere que el consumo de pitaya se asoció con una menor peroxidación lipídica. Este estudo teve como objetivo analisar as características físico-químicas deste fruto, avaliar sua composição centesimal e efeito in vivo sobre o estresse oxidativo em camundongos swiss diabéticas. Polpa, sementes e casca foram isoladas e utilizadas para análise da composição físico-química e centesimal. A DM foi induzida em camundongos suíços fêmeas por injeção intraperitoneal de Aloxano Monohidratado. Foram formados grupos com 6 animais: saudáveis (SAUD), não tratados A e B (DNT A e DNT B), metformina (MET) e grupos alimentados com 20 (PIT20), 40 (PIT40), 200 (PIT200) e 400 (PIT400) mg de extrato polpa com sementes/kg de peso corporal dos animais, por gavagem, por 28 dias. O conteúdo hepático das espécies reativas ao ácido tiobarbitúrico (TBARS) foi quantificado pelo Malondialdeído (MDA), para avaliar a peroxidação lipídica. A composição da pitaia foi semelhante à relatada anteriormente. Polpa e casca apresentaram conteúdo lipídico insignificante, presença expressiva de carboidratos e fibras em todas as amostras, principalmente na casca. O menor MDA foi observado em PIT400, PIT200 e PIT40, em comparação com DNT A e B e MET. Os resultados foram dose-dependentes, sugerindo que o consumo de pitaia estava associado a menor peroxidação lipídica.
