HTLV-1: genetic and epigenetic factors associated with the development of adult t-cell leukemia/lymphoma

Resumo: Objective: Identify possible genetic and epigenetic factors associated with the development of adult T-cell leukemia/lymphoma. Methodology: This is a work based on the integrative review of the literature. The search for articles was performed using the descriptors indexed in the DeCS (Descriptors in Health Sciences) and MeSH (Medical Subject Headings), articles published in the last 5 years were included. Results and Discussion: The carcinogenesis process that culminates in the progression of ATLL has several phases over a long period of time. The beginning of the neoplastic process is related to the expression of viral oncoprotein Tax, which induces replication and initiation of the clonal expansion process. HBZ oncoprotein and HBZ RNA are continuously transcribed into neoplastic cells and influence the process of cell proliferation and viral persistence. In addition to genetic factors that are generally mediated by Tax and HBZ proteins, epigenetic changes are also required, such as histone acetylation and DNA methylation. Conclusion: it is then concluded that genetic factors such as Tax, HBZ, FYN, PLCG1, CD247, DLG1, VAV1, ERC1, PRKCQ, CARD11, RELA, IKBKB, TET1, TET2, MLL2, MLL3 DNMT1 e IRF4. And epigenetic pathways that act on DNA methylation and histone acetylation are involved in the HTLV-1-mediated carcinogenesis process.

Metodología: Se trata de un trabajo basado en la revisión integradora de la literatura. La búsqueda de artículos se realizó utilizando los descriptores indexados en el DeCS (Descriptores en Ciencias de la Salud) y MeSH (Medical Subject Headings), se incluyeron artículos publicados en los últimos 5 años. Resultados y Discusión: El proceso de carcinogénesis que culmina en la progresión de ATLL tiene varias fases durante un largo período de tiempo. El inicio del proceso neoplásico está relacionado con la expresión de la oncoproteína viral Tax, que induce la replicación e inicio del proceso de expansión clonal. La oncoproteína HBZ y el ARN HBZ se transcriben continuamente en células neoplásicas e influyen en el proceso de proliferación celular y persistencia viral. Además de los factores genéticos que generalmente están mediados por las proteínas Tax y HBZ, también se requieren cambios epigenéticos, como la acetilación de histonas y la metilación del ADN. Conclusión: luego se concluye que factores genéticos como el Tax, HBZ, FYN, PLCG1, CD247, DLG1, VAV1, ERC1, PRKCQ, CARD11, RELA, IKBKB, TET1, TET2, MLL2, MLL3 DNMT1 e IRF4. Y las vías epigenéticas que actúan sobre la metilación del ADN y la acetilación de histonas están involucradas en el proceso de carcinogénesis mediada por HTLV-1.Objetivo: Identificar possíveis fatores genéticos e epigenéticos associados ao desenvolvimento da leucemia/linfoma de células T do adulto. Metodologia: Trata-se de trabalho baseado na revisão integrativa da literatura. A busca por artigos foi realizada mediante o uso dos descritores indexados no DeCS (Descritores em Ciências de saúde) e MeSH (Medical Subject Headings), foram incluídos artigos publicados nos últimos 5 anos. Resultados e Discussão: O processo de carcinogênese que culmina na progressão a ATLL possui várias fases em um longo período de tempo. O início do processo neoplásico está relacionado à expressão da oncoproteína viral Tax, que induz a replicação e início do processo de expansão clonal. A oncoproteína HBZ e o RNA HBZ são continuamente transcritos nas células neoplásicas e influenciam no processo de proliferação celular e persistência viral. Além de fatores genéticos que geralmente são mediadas pelas proteínas Tax e HBZ também é necessário alterações epigenéticas, como a acetilação de histonas e metilação do DNA. Conclusão: conclui-se então que fatores genéticos como o Tax, HBZ, FYN, PLCG1, CD247, DLG1, VAV1, ERC1, PRKCQ, CARD11, RELA, IKBKB, TET1, TET2, MLL2, MLL3 DNMT1 e IRF4.
