Evaluation of antigens by enzyme immunoassay ELISA during experimental infection in goats with Corynebacterium pseudotuberculosis

Resumo: | Corynebacterium pseudotuberculosis is the etiologic agent of caseous lymphadenitis (CL), a chronic infectious disease that affects sheep and goats. Numerous serological tests have been developed to detect the disease, being the Enzyme Immunoassay (ELISA) one of the most used due to its advantages of sensitivity and specificity. The present study aimed to evaluate the reactivity of a panel of Corynebacterium pseudotuberculosis antigens in goats experimentally infected with non-forming and biofilm-forming strains. Eighteen Canindé goats were used: group 1 - negative control animals, group 2 - goats infected with non-biofilm-forming bacteria and group 3 - goats infected with a biofilm-forming strain. Recombinant antigens CP40 and PLD, membrane fraction antigens (F3) and C. pseudotuberculosis secreted antigens (BHI) were tested. The sensitivity and specificity of the indirect ELISA tests for all antigens were 100%. However, the optical density (OD) values recorded in the BHI ELISA were lower than those found in the other ELISA tests, suggesting that specific antigens may have better sensitivity and specificity in the diagnosis of CL. |

Se han desarrollado numerosas pruebas serológicas para detectar la enfermedad, siendo el Inmunoensayo Enzimático (ELISA) una de las más utilizadas por sus ventajas de sensibilidad y especificidad. El presente estudio tuvo como objetivo evaluar la reactividad de un panel de antígenos de Corynebacterium pseudotuberculosis en cabras experimentalmente infectadas con cepas no formadoras y formadoras de biopelículas. Se utilizaron dieciocho cabras Canindé: grupo 1 - animales de control negativo, grupo 2 - cabras infectadas con bacterias no formadoras de biopelículas y grupo 3 - cabras infectadas con una cepa formadora de biopelículas. Se analizaron los antígenos recombinantes CP40 y PLD, los antígenos de fracción de membrana (F3) y los antígenos secretados por C. pseudotuberculosis (BHI). La sensibilidad y especificidad de las pruebas ELISA indirectas para todos los antígenos fue del 100%. Sin embargo, los valores de densidad óptica (OD) registrados en el BHI ELISA fueron más bajos que los encontrados en las otras pruebas de ELISA, lo que sugiere que los antígenos específicos pueden tener una mejor sensibilidad y especificidad en el diagnóstico de CL.Corynebacterium pseudotuberculosis é o agente etiológico da linfadenite caseosa (LC), uma doença infecciosa crônica que afeta ovinos e caprinos. O presente estudo teve como objetivo avaliar a reatividade de um painel de antígenos de Corynebacterium pseudotuberculosis em caprinos experimentalmente infectados com linhagens não formadoras e formadoras de biofilme. Foram utilizados 18 caprinos da raça Canindé sendo: grupo 1 - animais controle negativo, grupo 2 - caprinos infectados com bactéria não formadora de biofilme e grupo 3 - caprinos infectados com linhagem formadora de biofilme. Foram testados os antígenos recombinantes CP40 e PLD, antígenos de fração de membrana (F3) e antígenos secretados de C. pseudotuberculosis (BHI). A sensibilidade e especificidade dos testes ELISA indireto para todos os antígenos foram de 100%.
