Vaccine design by immunoinformatics against HCV: Chemical characterization and prediction of T-cell epitopes

Resumo: | Hepatitis C is a disease that affects the liver causing its inflammation, reaching thousands of people every year, however currently there are no vaccines for the virus responsible for the disease, HCV, although studies are already carried out on its feasibility. Therefore, reverse vaccinology can be of great help in detecting promising targets for use as a vaccine antigen and concomitantly formulating an effective vaccine. The respective work aims to obtain relevant data for the design of promising vaccines against HCV through the use of bioinformatics tools. Therefore, after the virus sequences were obtained from the National Center for Biotechnological Information (NCBI), analyzes of the antigenic potential were carried out using VaxiJen v.2.0. and from ANTIGENpro, together with AlgPred, the allergenic potential was verified, soon after the proteins were verified and selected, they would proceed to the next steps where they were submitted for the characterization of the physicochemical properties, through and the prediction of T cell epitopes was performed on NetTepi. Among the results obtained, it can be noted that the E2 protein was the only one to demonstrate allergenicity, whereas the E1 protein did not present antigenic potential and the other proteins were classified as unstable, following only the NS4a protein for prediction of T cell epitopes. |

Por lo tanto, la vacunación inversa puede ser de gran ayuda para detectar dianas prometedoras para su uso como antígeno de vacuna y, concomitantemente, formular una vacuna eficaz. El trabajo respectivo tiene como objetivo obtener datos relevantes para el diseño de vacunas prometedoras contra el VHC mediante el uso de herramientas bioinformáticas. Por lo tanto, después de que se obtuvieron las secuencias de virus del Centro Nacional de Información Biotecnológica (NCBI), se realizaron análisis del potencial antigénico utilizando VaxiJen v.2.0. y desde ANTIGENpro, en conjunto con AlgPred, se verificó el potencial alergénico, luego de verificadas y seleccionadas las proteínas, se pasaría a los siguientes pasos donde se sometían a la caracterización de las propiedades fisicoquímicas, mediante y la predicción de epítopos de células T se realizó en NetTepi. Entre los resultados obtenidos, se puede observar que la proteína E2 fue la única que demostró alergenicidad, mientras que la proteína E1 no presentó potencial antigénico y las demás proteínas se clasificaron como inestables, siguiendo solo a la proteína NS4a para la predicción de epítopos de células T.A hepatite C é uma enfermidade que atinge o fígado causando sua inflamação, alcançando milhares de pessoas todos os anos, contudo atualmente ainda não existem vacinas para o vírus responsável pela doença, o HCV, embora estudos já sejam realizados sobre a viabilidade. Assim sendo, a vacinologia reversa pode ser de grande auxílio para detecção de alvos promissores para utilização como antígeno vacinal e concomitantemente formulação de uma vacina eficaz. O respectivo trabalho tem como objetivo obter dados relevantes para o desenho de vacinas promissoras contra o HCV através da utilização de ferramentas de bioinformática. Para tanto, após as sequências do vírus serem obtidas no Centro Nacional de Informação Biotecnológica (NCBI), foram realizadas as análises do potencial antigênico através do VaxiJen v.2.0. e do ANTIGENpro, juntamente foi verificado o potencial alergênico com o AlgPred, logo após as proteínas serem selecionadas, seguiriam para as próximas etapas onde foram submetidas para a caracterização das propriedades físico-químicas, através do ProtParam e a predição de epítopos de células T foi realizada no NetTepi.
