Germination and seedling growth in vitro of Pilosocereus pachycladus Ritter (Facheiro) submitted to different culture media

Resumo: The productive versatility of the facheiro associated with the natural availability of the species causes an exploration without planning. Consequently, few studies have been carried out on the conservation and maintenance of the species, mainly in the Northeast Region. The aim of the present study was to evaluate the effect of the simplified medium and the alternative gelling agent, corn starch, on the in vitro germination of Pilosocereus pachycladus. The seeds were disinfected and inoculated in a culture medium containing commercial fertilizers, supplemented with sucrose, and gelled with agar and corn starch. The treatments: complete salts of Calcinit® and Kristalon® and gelled with agar - CKAG; half of the Calcinit® and Kristalon® salts and gelled with agar - MCKAG; complete salts of Calcinit® and Kristalon® and gelled with corn starch - CKAM; half of the Calcinit® and Kristalon® salts and gelled with corn starch - MCKAM. The CKAG medium promoted a lower percentage of contamination by 20%. And the CKAM and MCKAM media obtained 67 and 75% for germination percentage, respectively. The highest averages for aerial part height were for the CKAM and CKAG media with 0.32 and 0.30 cm, respectively. And for the length of the root, the highest value was for the CKAG medium with 0.65 cm. There was no significant effect on seedling acclimatization for treatments. In view of the results obtained, it is suggested the use of corn starch in culture media as an excellent alternative for in vitro germination of the P. pachycladus, providing cost reduction and a satisfactory germination percentage.

En consecuencia, se han realizado pocos estudios sobre la conservación y el mantenimiento de la especie, principalmente en la región noreste. El objetivo del presente estudio fue evaluar el efecto del medio simplificado y el agente gelificante alternativo, el almidón de maíz, sobre la germinación in vitro de Pilosocereus pachycladus. Las semillas fueron desinfectadas e inoculadas en un medio de cultivo que contenía fertilizantes comerciales, suplementados con sacarosa y gelificados con agar y almidón de maíz. Los tratamientos: sales completas de Calcinit® y Kristalon® y gelificadas con agar - CKAG; la mitad de las sales de Calcinit® y Kristalon® y gelificadas con agar - MCKAG; sales completas de Calcinit® y Kristalon® y gelificadas con almidón de maíz - CKAM; mitad de las sales de Calcinit® y Kristalon® y gelificado con almidón de maíz - MCKAM. El medio CKAG promovió un menor porcentaje de contaminación del 20%. Y los medios CKAM y MCKAM obtuvieron 67 y 75% para el porcentaje de germinación, respectivamente. Los promedios más altos para la altura de la parte aérea fueron para los medios CKAM y CKAG con 0.32 y 0.30 cm, respectivamente. Y para la longitud de la raíz, el valor más alto fue para el medio CKAG con 0,65 cm. No hubo un efecto significativo en la aclimatación de plántulas para los tratamientos. En vista de los resultados obtenidos, se sugiere el uso de almidón de maíz en medios de cultivo como una excelente alternativa en la germinación in vitro de las P. pachycladus, proporcionando una reducción de costos y un porcentaje de germinación satisfactorio.A versatilidade produtiva do facheiro associada a disponibilidade natural da espécie ocasionam uma exploração sem planejamento. Consequentemente, poucos estudos tem sido desenvolvidos sobre a conservação e manutenção da espécie, principalmente na Região Nordeste.  O objetivo do presente estudo foi avaliar o efeito do meio simplificado e do agente gelificante alternativo, amido de milho, na germinação in vitro de Pilosocereus pachycladus. As sementes foram desinfestadas e inoculadas em meio de cultivo contendo fertilizantes comerciais, suplementados com sacarose, e gelificados com ágar e amido de milho. Os tratamentos: sais completo de Calcinit® e Kristalon® e gelificado com ágar - CKAG; metade dos sais de Calcinit® e Kristalon® e gelificado com ágar – MCKAG; sais completo de Calcinit® e Kristalon® e gelificado com amido de milho - CKAM; metade dos sais de Calcinit® e Kristalon® e gelificado com amido de milho - MCKAM. O meio CKAG promoveu menor percentual de contaminação 20%. E os meios CKAM e MCKAM obtiveram 67 e 75% para percentual de germinação respectivamente. As maiores médias para altura da parte aérea foi para os meios CKAM e CKAG com 0,32 e 0,30 cm respectivamente. E para o comprimento da raiz o maior valor foi para o meio CKAG com 0,65 cm. Não houve efeito significativo na aclimatação das plântulas para os tratamentos.
