Systematic review of the clinical and experimental use of pregabalin and its effects on the male reproductive system and the endocrine system

Resumo: | Pregabalin is a drug commonly used in psychiatry, neurology, and primary health care. It is an anticonvulsant widely used in the treatment of epilepsy, anxiety, fibromyalgia and neuropathic pain. It works by binding to voltage-gated calcium channels, exerting analgesic and anxiolytic activity. It has GABA-like properties and is involved in the dopaminergic reward system. The objective of this systematic review was to describe the status of current knowledge in relation to the topic, highlighting possible paths for future research, guiding other researchers and directing new publications with objectives correlated to the current diagnosis. In the present systematic review, we used original and comprehensive, open access scientific research, carried out between 2016 and 2021, related to the drug Pregabalin, highlighting its relationship with the testes and the Endocrine System in male rats. In order to map the scientific production, an exploratory-descriptive research was carried out, through a systematic analysis. The main collection of the Web of Science (Clarivate Analytics) was used as a search base. Studies have shown that pregabalin affects FSH, LH and testosterone levels, causing changes in spermatogenesis. Pregabalin causes significant changes in hormone levels, negatively impacting the production of healthy sperm. Psychotropic drugs, particularly pregabalin, can also reduce sperm quality, decreasing patients fertility potential. |

Es un anticonvulsivo ampliamente utilizado en el tratamiento de la epilepsia, la ansiedad, la fibromialgia y el dolor neuropático. Actúa uniéndose a los canales de calcio dependientes de voltaje, ejerciendo actividad analgésica y ansiolítica. Tiene propiedades similares al GABA y está involucrado en el sistema de recompensa dopaminérgico. El objetivo de esta revisión sistemática fue describir el estado del conocimiento actual en relación al tema, destacando posibles caminos para futuras investigaciones, orientando a otros investigadores y orientando nuevas publicaciones con objetivos correlacionados al diagnóstico actual. En la presente revisión sistemática, utilizamos investigaciones científicas originales, integrales y de acceso abierto, realizadas entre 2016 y 2021, relacionadas con el fármaco Pregabalina, destacando su relación con los testículos y el Sistema Endocrino en ratas macho. Para mapear la producción científica, se realizó una investigación exploratoria-descriptiva, a través de un análisis sistemático. Se utilizó como base de búsqueda la colección principal de la Web of Science (Clarivate Analytics). Los estudios han demostrado que la pregabalina afecta los niveles de FSH, LH y testosterona, provocando cambios en la espermatogénesis. La pregabalina provoca cambios significativos en los niveles hormonales, lo que afecta negativamente la producción de espermatozoides sanos. Los fármacos psicotrópicos, en particular la pregabalina, también pueden reducir la calidad del esperma, disminuyendo el potencial de fertilidad de los pacientes.A pregabalina é uma droga comumente usada na psiquiatria, neurologia e em cuidados primários de saúde.
