Synthesis and characterization of O-Carboxymethylchitosan as an alternative to the use of Hyaluronic Acid

Resumo: Hyaluronic acid (HA) has been an important component in several fields, such as dermal filling, tissue engineering, treatment of the vascular and cartilaginous region, and its application is limited because its excessive use can promote a reduction in cell adhesion, due to its presence of negative charges, characteristic of this material. To solve this limitation, a cationic and anionic material can be used, such as O-Carboxymethylchitosan (O-CMQ), which has a similar chemical structure and similar properties to HA. Therefore, this work aimed to determine the conditions for the synthesis and characterization of O-Carboxymethylchitosan (O-CMQ), and to evaluate its application as an alternative to hyaluronic acid. From the chitosan produced and characterized, the synthesis of O-CMQ was carried out in the proportions 1:1, 1:2 and 1.5:2 (m/v), followed by the characterizations through Visual Analysis, Optical Microscopy (OM), Fourier Transform Infrared Spectroscopy (FTIR), Hydrogen Potential (pH) and Conductivity, Thermogravimetry Analysis (TGA) and Differential Scanning Calorimetry (DSC). It was concluded that the synthesis methodology was efficient for the production of O-Carboxymethylchitosan with different properties. The comparison with hyaluronic acid confirmed the chemical, physical and biological similarities between the samples, being promising for application as a substitute for this material.

Para solucionar esta limitación, se puede utilizar un material catiónico y aniónico, como el O-Carboximetilquitosano (O-CMQ), que tiene una estructura química similar y propiedades similares al HA. Por tanto, este trabajo tuvo como objetivo determinar las condiciones de síntesis y caracterización del O-Carboximetilquitosano (O-CMQ), y evaluar su aplicación como alternativa al ácido hialurónico. A partir del quitosano producido y caracterizado, se realizó la síntesis de O-CMQ en las proporciones 1:1, 1:2 y 1.5:2 (m/v), seguido de las caracterizaciones por Análisis Visual, Microscopía Óptica (OM), Espectroscopia Infrarroja por Transformada de Fourier (FTIR), Potencial de Hidrógeno (pH) y Conductivimetría, Análisis Termogravimétrico (TGA) y Calorimetría Exploratoria Diferencial (DSC). Se concluyó que la metodología de síntesis fue eficiente para la producción de O-Carboximetilquitosano con diferentes propiedades. La comparación con el ácido hialurónico confirmó las similitudes químicas, físicas y biológicas entre las muestras, siendo prometedora su aplicación como sustituto de este material.O ácido Hialurônico (AH) tem sido um componente importante em diversos campos, como preenchimento dérmico, engenharia de tecidos, tratamento da região vascular e cartilaginosa, tendo sua aplicação limitada pois o uso em excesso pode promover a redução da adesão celular, devido à presença de cargas negativas, característico deste material. Para solucionar esta limitação, pode ser empregado um material catiônico e aniônico, tal como a O-Carboximetilquitosana (O-CMQ), que possui uma estrutura química similar e propriedades análogas ao AH. Portanto, esse trabalho teve como objetivo a determinação das condições de síntese e caracterização da O-Carboximetilquitosana (O-CMQ), e avaliação da aplicação como alternativa ao ácido hialurônico. A partir da quitosana produzida e caracterizada foi realizada a síntese da O-CMQ nas proporções 1:1, 1:2 e 1,5:2 (m/v), seguindo das caracterizações através de Análise Visual, Microscopia Óptica (MO), Espectroscopia no Infravermelho por Transformada de Fourier (FTIR), Potencial Hidrogeniônico (pH) e Condutivimetria, Análise Termogravimétrica (TGA) e Calorimetria Diferencial Exploratória (DSC). Concluiu-se que a metodologia de síntese foi eficiente para a produção da O-Carboximetilquitosana com diferentes propriedades.
