Resumo: The Fusarium genus is among the pathogens that cause onychomycosis (OM), and is included among the non-dermatophyte filamentous fungi, with the Fusarium solani and Fusarium oxysporum species standing out. Among the therapeutic options for OM, Photodynamic Therapy (PDT) stands out, which, associated with Curcumin (CUR), is an interesting proposal in the antifungal context. The aim was to standardize and evaluate the antifungal activity of PDT+CUR on clinical isolates of Fusarium spp. The CUR was supplied by the UEM Photodynamic Research Center, and the Fusarium strains were obtained from samples from the Clinical Analysis Teaching and Research Laboratory, approved by the ethics committee (nº2.748.843). Blue LED (440nm to 460nm) was used for irradiation, and CUR was used at concentrations of 50-0.09µmol/L. The inoculum of the isolates was standardized at a concentration of 105 CFU/mL. After incubation for 10, 20 and 30 minutes in the dark with CUR, the plates were irradiated at doses of 15, 30 and 45 J/cm2. The antifungal action of PDT+CUR was assessed qualitatively and quantitatively. The results were analyzed using the GraphPad Prism 5.0® software, with One-Way Analysis of Variance (ANOVA) and Tukey's test. The best antifungal combination of incubation time and fluence of PDT+CUR was 10 minutes for pre-irradiation and 30 J/cm2 irradiation. These conditions were able to inactivate 75% of the clinical isolates tested from a concentration of 0.78µmol/L. The results of this study show that PDT+CUR was fungicidal and promising, encouraging further research in the field of medical mycology.

Dentre as opções terapêuticas para OM, destaca-se a Terapia Fotodinâmica (TFD), que associada à Curcumina (CUR), mostra-se como uma proposta interessante no contexto antifúngico. O objetivo foi padronizar e avaliar a atividade antifúngica da TFD+CUR sobre isolados clínicos de Fusarium spp. A CUR foi fornecida pelo Núcleo de Pesquisa Fotodinâmicos UEM, as cepas de Fusarium, foram obtidas de amostras do Laboratório de Ensino e Pesquisa em Análises Clínicas, aprovado pelo comitê de ética (nº2.748.843). Para irradiação utilizou-se LED azul (440nm a 460nm), e a CUR foi utilizada nas concentrações de 50-0,09µmol/L. O inóculo dos isolados foi padronizado na concentração 105 UFC/mL. Após incubação de 10, 20 e 30 minutos no escuro com CUR, as placas foram irradiadas em doses: 15, 30 e 45 J/cm2. A ação antifúngica da TFD+CUR foi avaliada qualitativamente e quantitativamente. Analisou-se os resultados pelo programa GraphPad Prism 5.0®, com teste One-Way Analysis of Variance (ANOVA) e Tukey. A melhor combinação antifúngica entre tempo de incubação e fluência da TFD+CUR foi de 10 minutos para pré-irradiação e irradiação de 30 J/cm2. Nessas condições foi capaz de inativar 75% dos isolados clínicos testados a partir da concentração de 0.78µmol/L. Os resultados deste estudo mostram que a TFD+CUR foi fungicida e promissora, incentivando outras pesquisas na área da micologia médica.  El género Fusarium se encuentra entre los patógenos causantes de onicomicosis (OM), y se incluye entre los hongos filamentosos no dermatofitos, destacando las especies Fusarium solani y Fusarium oxysporum. Entre las opciones terapéuticas para la OM destaca la Terapia Fotodinámica (TFD) que, asociada a la Curcumina (CUR), constituye una interesante propuesta en el contexto antifúngico. El objetivo fue estandarizar y evaluar la actividad antifúngica de la TFD+CUR sobre aislados clínicos de Fusarium spp. El CUR fue suministrado por el Centro de Investigación Fotodinámica de la UEM, y las cepas de Fusarium se obtuvieron a partir de muestras del Laboratorio de Docencia e Investigación en Análisis Clínicos, aprobadas por el comité ético (nº2.748.843). Para la irradiación se utilizó LED azul (440nm a 460nm) y CUR a concentraciones de 50-0,09µmol/L. El inóculo de los aislados se estandarizó a una concentración de 105 UFC/mL. Tras incubar durante 10, 20 y 30 minutos en la oscuridad con CUR, las placas se irradiaron a dosis de 15, 30 y 45 J/cm2. La acción antifúngica de TFD+CUR se evaluó cualitativa y cuantitativamente. Los resultados se analizaron mediante el programa GraphPad Prism 5.0®, con análisis de varianza unidireccional (ANOVA) y la prueba de Tukey. La mejor combinación antifúngica de tiempo de incubación y fluencia de TFD+CUR fue de 10 minutos para la pre-irradiación y 30 J/cm2 de irradiación. Estas condiciones fueron capaces de inactivar el 75% de los aislados clínicos ensayados a partir de una concentración de 0,78µmol/L.
