Maternal exposure to glyphosate-based herbicide causes long-term changes in vascular relaxation from offspring adult rats

Resumo: This study aimed to investigate whether exposure to glyphosate-based herbicide during the gestational phase promotes long-term effects on the vasomotor function of the thoracic aorta and the possible involvement of oxidative stress and cyclooxygenase II in these alterations. For this, in a isolated organ bath, the endothelium-dependent and independent relaxation of aortic rings of rats from mothers treated with glyphosate in the maternal phase (O-GHB) and tap water the control group (O-CON) were evaluated. Pharmacological tools such as L-NAME, TEMPOL and Indomethacin were used for investigate possible mechanisms. As a result, an impairment in vascular relaxation related to an endothelial dysfunction in O-GHB rats was observed. This endothelial dysfunction was reversed when the aortic rings were in the presence of superoxide dismutase and COX-2 mimetic, supporting the hypothesis that adverse effects during the intrauterine phase, such as oxidative stress caused by glyphosate, can lead to late changes in the function of some organs, in the case studied here, in vascular reactivity in the aorta.

Para ello, se evaluó la relajación dependiente e independiente del endotelio de los anillos aórticos de ratas de madres tratadas con glifosato en fase materna (O-GHB) y control (O-CON) en un baño de órganos aislados. Para analisar posibles mecanismos fármacos como L -NAME, TEMPOL e Indometacina fueron usados. Como resultado, se observó una alteración en la relajación vascular relacionada con una disfunción endotelial en ratas O-GHB. Esta disfunción endotelial se revirtió cuando los anillos aórticos estaban en presencia de superóxido dismutasa y mimético de COX-2, apoyando la hipótesis de que los efectos adversos durante la fase intrauterina, como el estrés oxidativo causado por el glifosato, pueden conducir a cambios tardíos en la función de algunos órganos, en el caso aquí estudiado, en reactividad vascular en la aorta.Este estudo se propôs a investigar se a exposição ao herbicida baseado em glifosato durante a fase gestacional promove efeitos em longo prazo sobre a função vasomotora da aorta torácica e o possível envolvimento de estresse oxidativo e da ciclooxigenase II nessas alterações. Para isso foram avaliadas em banho de órgãos isolados, o relaxamento dependente e independente do endotélio de anéis aórticos de ratos provenientes de mães tratadas com glifosato na fase materna (O-GHB) e controle (O-CON) e para os possíveis mecanismos se utilizou ferramentas farmacológicas como L-NAME, TEMPOL e Indometacina. Como resultados foi observado um prejuízo no relaxamento vascular relacionado a uma disfunção endotelial nos ratos O-GHB.
