Evaluation of liposomal particle size in different assays

Resumo: Liposomes composed of rice (RL) and soy (SL) lecithins were produced by reverse phase evaporation, using as a solvent for solubilization of rice and soy lecithins the Chloroform P.A. With this, the objective was to perform different assays to evaluate the average size and polydispersity index of liposomes with and without the incorporation of the phenolic compound extracted from Spirulina LEB - 18. The pure liposomes were characterized in size distribution, polydispersity index, and ζ-potential. Liposomes in different assays, on the other hand: A) Liposomes samples with and without phenolic extracts of microalgae were freeze-dried in a freeze-dryer (model LIOB 003, Biosan), were subjected to solubilization in Ultrapure water for characterization. B) Liposome samples with and without phenolic extracts of microalgae were made by centrifugation at 10,000 g for 30 minutes. C) Liposomes samples with and without phenolic extracts of microalgae were submitted to centrifugation at 10,000 g for 30 min and filtration through a membrane filter of 0.22 (Filter For Syringe 0.22 a Kasvi) and 0.45 µm pore size (Membrane Millipore). Then these assays were subjected to size and polydispersity evaluation. Therefore, it was verified that assay C obtained the best sizes. With the results obtained, it is possible to emphasize the role of liposomes as an essential tool for enhancing the antioxidant profile of phenolic extracts of Spirulina sp. LEB-18.

LEB-18.Los liposomas compuestos por lecitina de arroz (RL) y lecitina de soja (SL) se produjeron por evaporación en fase inversa, utilizando como disolvente para la solubilización de las lecitinas de arroz y soja el cloroformo P.A . Con esto el objetivo fue realizar diferentes ensayos para evaluar el tamaño medio y el índice de polidispersidad de los liposomas con y sin la incorporación del compuesto fenólico extraído de la Spirulina LEB - 18. Los liposomas puros se caracterizaron en términos de distribución de tamaño, índice de polidispersidad y potencial ζ. Por otro lado, los liposomas en diferentes ensayos: A) Las muestras de liposomas con y sin extractos fenólicos de microalgas fueron liofilizadas, en liofilizador (modelo LIOB 003, Biosan), fueron sometidas a solubilización en agua ultrapura para su caracterización. B) Las muestras de liposomas con y sin extractos fenólicos de microalgas se realizaron por centrifugación a 10.000 g durante 30 minutos. C) Las muestras de liposomas con y sin extractos fenólicos de microalgas se sometieron a centrifugación a 10.000 g durante 30 min y a filtración en filtro de membrana de 0,22 (Filtro Para Jeringa 0,22 a Kasvi) y 0,45 µm de tamaño de poro (Membrana Milipore). Por lo tanto, estos ensayos se sometieron a la evaluación del tamaño y la polidispersidad. Por lo tanto, se verificó que el ensayo C obtuvo mejores tamaños, con los resultados obtenidos se puede destacar el papel de los liposomas como una herramienta importante para potenciar el perfil antioxidante de los extractos fenólicos de Spirulina sp. LEB-18.Lipossomas compostos por lecitinas de arroz (RL) e soja (SL) foram produzidos por evaporação de fase reversa, utilizando como solvente para solubilização das lecitinas de arroz e soja o Clorofórmio P.A. Com isso o objetivo foi realizar diferentes ensaios para avaliar o tamanho médio e índice de polidispersão de lipossomas com e sem a incorporação do composto fenólico extraído de Spirulina LEB – 18. Os lipossomas puros foram caracterizados em termos de distribuição de tamanho, índice de polidispersidade, e ζ-potencial. Já os Lipossomas em diferentes ensaios: A) As amostras de lipossomas com e sem extratos fenolicos de microalgas foram liofilizadas, em liofilizador (modelo LIOB 003, Biosan), foram submetidas a solubilização em água Ultrapura para a caracterização. B) As amostras de lipossomas com e sem extratos fenolicos de microalgas foram executadas através da centrifugação a 10.000 g por 30 minutos. C) As amostras de lipossomas com e sem extratos fenolicos de microalgas foram submetidas a centrifugação a 10.000 g por 30 min e filtração em filtro de membranas de 0.22 (Filtro Para Seringa 0.22 um Kasvi) e 0.45 µm tamanho do Poro (Membrana Milipore). Logo, estes ensaios foram submetidos a avaliação dos tamanhos e polidispersão. Portanto, foi verificado que o ensaio C obteve-se melhores tamanhos, com os resultados obtidos é possível enfatizar o papel dos lipossomas como importante ferramenta para potencializar o perfil antioxidante de extratos fenólicos de Spirulina sp.
