Eficácia de níveis de um blend de aditivos sobre o desempenho e a saúde intestinal de frangos de corte desafiados experimentalmente

Resumo: | The maintenance of good intestinal integrity and health are of fundamental importance so that production animals can be able to express their genetic potential. In order to measure the impact of different variables involved in the context of broiler production, it is necessary to apply experimental protocols that are capable of simulating the reality of the field, allowing the accurate evaluation of food additives. In this sense, we aim to evaluate the performance, morphology, and regenerative capacity of the intestinal mucosa, as well as to study intestinal microbiota responses and the immune response of birds fed with a blend of additives composed of zinc proteinate, hydrolyzed yeast and encapsulated sodium butyrate associated or not with the use of antibiotic growth promoter in broilers submitted or not to an enteric challenge proposal. This study was carried out in the experimental aviary of the Federal University of Paraná – Palotina campus. A total of 3,936 male broiler chicks (Ross AP95®) were used and the birds were housed on shavings bedding, with water and feed ad libitum and divided into a 6 x 2 factorial scheme (Negative control (NC), Positive control (PC); PC + 1.0kg of Blend of additives/ton of feed; NC + 0.5kg; NC + 1.0kg; NC + 1.5kg of Blend of additives/ton of feed Vs with and without experimental enteric challenge), with 8 replications per treatment and 41 birds each. At 14 days of age, birds in the challenged groups were inoculated into the coccidiosis vaccine pool containing 20 times the dose recommended by the manufacturer. Two days later (16th day), the challenged birds received an inoculum containing Escherichia coli (ATCC® 8739™) with a calculated concentration of 109 CFU/bird applied directly to the flock. The evaluation of body weight, average feed intake, weight gain, and feed conversion were performed weekly. The degree of warpage was evaluated at 28, 35 and 42 days of age. Fragments of intestinal segments were collected at 19 days of age after euthanasia procedure of two birds/repeat for evaluation of intestinal morphometry and analysis of the proliferative capacity of intestinal cells. Intestinal mucosal permeability was measured by assessing serum levels of fluorescein isothiocyanate (FITC-d). Cecal excreta samples were collected at 42 days of age from 7 birds per treatment to determine the short chain fatty acid (SCFA) profile. For the calculation of carcass yield, cuts and abdominal fat deposition, at 42 days, 60 birds were slaughtered per treatment. Statistical procedures were performed using analysis of variance (ANOVA) using the General Lineal Model (GLM) using the SAS statistical program and, when significant, the treatment means were compared using Tukey's test (p<0.05). The results of this evaluation indicate that the enteritis induction protocol proposed in this study was effective, with compromised performance (p<0.05) in relation to the group of birds that were not experimentally exposed to the challenge. The birds that did not receive AGP in the diets and were supplemented with 0.5kg and 1.5kg of inclusion of the blend of feed additives showed similar results for WG to those birds that received diets with AGP. When considering the post-challenge period and the total rearing period, the birds supplemented with 1.5kg of the feed additive blend were those that obtained the best (p<0.05) feed conversion rate, as well as the birds supplemented with 1.0kg of the blend associated with AGP showed higher (p<0.05) weight gain in relation to the other diets. The blend of food additives composed of zinc proteinate, hydrolyzed yeast and encapsulated sodium butyrate used in the diets of broilers from 1 to 42 days of age submitted or not to the enteric challenge proposed in this experiment provided results similar or even superior to those when using AGP. Thus, this Blend can be indicated to act as an alternative or together with AGP in the creation of broilers. |

Para que seja possível dimensionar o impacto de diferentes variáveis envolvidas no contexto da produção de frangos de corte há a necessidade de aplicação de protocolos experimentais que sejam capazes de simular a realidade de campo possibilitando a avaliação precisa dos aditivos alimentares. Nesse sentido, objetivamos avaliar o desempenho, a morfologia e a capacidade regenerativa da mucosa intestinal bem como estudar respostas da microbiota intestinal e a resposta imune das aves alimentadas com um blend de aditivos composto por proteinato de zinco, levedura hidrolisada e butirato de sódio encapsulado (BSE) associado ou não ao uso de antibiótico melhorador de desempenho (AMD) de frangos de corte submetidos ou não a uma proposta de desafio entérico. Este estudo foi realizado no aviário experimental da Universidade Federal do Paraná – campus Palotina. Foram utilizados 3.936 pintos de corte, machos (Ross AP95®) sendo as aves alojadas sobre cama de maravalha, com água e ração ad libitum e divididas em um esquema fatorial 6 x 2 (Controle negativo (CN); Controle positivo (CP); CP + 1.0kg de Blend de aditivos/tonelada de ração; CN + 0.5kg; CN + 1.0kg; CN + 1.5kg de Blend de aditivos/tonelada de ração Vs com e sem desafio entérico experimental), com 8 repetições por tratamento e 41 aves cada. Aos 14 dias de idade, as aves dos grupos desafiados receberam inoculação no inglúvio de vacina para coccidiose contendo 20 vezes a dose recomendada pelo fabricante. Dois dias após (16º dia), as aves desafiadas receberam um inóculo contendo Escherichia coli (ATCC® 8739™) com uma concentração calculada de 109 UFC/ave aplicado diretamente no inglúvio. A avaliação do peso corporal (PC), consumo médio de ração (CMR), ganho de peso (GP) e conversão alimentar (CA) foram realizadas semanalmente. O grau de empenamento foi avaliado aos 28, 35 e 42 dias de idade. Fragmentos dos segmentos intestinais foram colhidos aos 19 dias de idade após procedimento de eutanásia de duas aves/repetição para avaliação da morfometria intestinal e análise da capacidade proliferativa das células intestinais. A permeabilidade da mucosa intestinal foi mensurada pela avaliação dos níveis séricos de isotiocianato de fluoresceína (FITC-d). Amostras de excretas cecais foram colhidas aos 42 dias de idade oriundas de 7 aves por tratamento para determinar o perfil de ácidos graxos de cadeia curta (AGCC). Para o cálculo do rendimento carcaça, cortes e deposição de gordura abdominal, aos 42 dias, foram abatidas 60 aves por tratamento. Os procedimentos estatísticos foram realizados através da análise de variância (ANOVA), do General Lineal Model (GLM) com auxílio do programa estatístico SAS e, quando significativas, as médias dos tratamentos foram comparadas pelo teste de Tukey (p<0,05). Os resultados desta avaliação indicam que o protocolo de indução de enterite proposto neste estudo foi eficaz, com o desempenho comprometido (p<0,05) em relação ao grupo de aves que não foi experimentalmente exposto ao desafio. As aves que não receberam AMD nas rações e foram suplementadas com 0,5kg e 1,5kg de inclusão do blend de aditivos alimentares apresentaram resultados similares para GP e CA em comparação com as aves que receberam dietas com AMD. Ao ser considerado o período pós desafio e o período total de criação, as aves suplementadas com 1,5kg do blend de aditivos alimentares foram aquelas que obtiveram melhor (p<0,05) CA, assim como as aves suplementadas com 1,0kg do blend associado ao AMD apresentaram maior (p<0,05) GP em relação as demais dietas. O blend de aditivos composto por proteinato de zinco, levedura hidrolisada e butirato de sódio encapsulado utilizado nas dietas de frangos de corte de 1 a 42 dias de idade submetidos ou não a desafio entérico proposto neste experimento proporcionou resultados de desempenho similares ou até mesmo superiores aqueles observadas com dietas suplementadas com AMD. The maintenance of good intestinal integrity and health are of fundamental importance so that production animals can be able to express their genetic potential. In order to measure the impact of different variables involved in the context of broiler production, it is necessary to apply experimental protocols that are capable of simulating the reality of the field, allowing the accurate evaluation of food additives. In this sense, we aim to evaluate the performance, morphology, and regenerative capacity of the intestinal mucosa, as well as to study intestinal microbiota responses and the immune response of birds fed with a blend of additives composed of zinc proteinate, hydrolyzed yeast and encapsulated sodium butyrate associated or not with the use of antibiotic growth promoter in broilers submitted or not to an enteric challenge proposal. This study was carried out in the experimental aviary of the Federal University of Paraná – Palotina campus. A total of 3,936 male broiler chicks (Ross AP95®) were used and the birds were housed on shavings bedding, with water and feed ad libitum and divided into a 6 x 2 factorial scheme (Negative control (NC), Positive control (PC); PC + 1.0kg of Blend of additives/ton of feed; NC + 0.5kg; NC + 1.0kg; NC + 1.5kg of Blend of additives/ton of feed Vs with and without experimental enteric challenge), with 8 replications per treatment and 41 birds each. At 14 days of age, birds in the challenged groups were inoculated into the coccidiosis vaccine pool containing 20 times the dose recommended by the manufacturer. Two days later (16th day), the challenged birds received an inoculum containing Escherichia coli (ATCC® 8739™) with a calculated concentration of 109 CFU/bird applied directly to the flock. The evaluation of body weight, average feed intake, weight gain, and feed conversion were performed weekly. The degree of warpage was evaluated at 28, 35 and 42 days of age. Fragments of intestinal segments were collected at 19 days of age after euthanasia procedure of two birds/repeat for evaluation of intestinal morphometry and analysis of the proliferative capacity of intestinal cells. Intestinal mucosal permeability was measured by assessing serum levels of fluorescein isothiocyanate (FITC-d). Cecal excreta samples were collected at 42 days of age from 7 birds per treatment to determine the short chain fatty acid (SCFA) profile. For the calculation of carcass yield, cuts and abdominal fat deposition, at 42 days, 60 birds were slaughtered per treatment. Statistical procedures were performed using analysis of variance (ANOVA) using the General Lineal Model (GLM) using the SAS statistical program and, when significant, the treatment means were compared using Tukey's test (p<0.05). The results of this evaluation indicate that the enteritis induction protocol proposed in this study was effective, with compromised performance (p<0.05) in relation to the group of birds that were not experimentally exposed to the challenge. The birds that did not receive AGP in the diets and were supplemented with 0.5kg and 1.5kg of inclusion of the blend of feed additives showed similar results for WG to those birds that received diets with AGP. When considering the post-challenge period and the total rearing period, the birds supplemented with 1.5kg of the feed additive blend were those that obtained the best (p<0.05) feed conversion rate, as well as the birds supplemented with 1.0kg of the blend associated with AGP showed higher (p<0.05) weight gain in relation to the other diets. The blend of food additives composed of zinc proteinate, hydrolyzed yeast and encapsulated sodium butyrate used in the diets of broilers from 1 to 42 days of age submitted or not to the enteric challenge proposed in this experiment provided results similar or even superior to those when using AGP. Thus, this Blend can be indicated to act as an alternative or together with AGP in the creation of broilers. |
