Uso de membrana reabsorvível de cortical bovina em defeitos críticos em calvária de ratos

Resumo: | Bone atrophy can result from etiologies as pathologies, trauma, surgical procedures and result of the physiological bone resorption following tooth loss. This fact has provided extensive studies on the process of bone repair. The objective of this study was to evaluate the efficacy of collagen absorbable membrane in critical defects in rat calvaria. Rattus norvegicus for that 36 were divided into control and treated groups. Animals were 8 mm diameter calvaria defects in their conducted specifically on the median sagittal suture in the frontal and parietal bones right and left. In the control group the defect was filled only by blood clot, while the treated group was filled with blood clot (GC), and covered with absorbable membrane (GCM) bovine collagen. The animals were euthanized at 7, 30 and 60 days postoperatively. The calvaria were removed and the excess material was removed, leaving only the cranial bones. The pieces were fixed in 10% formalin, followed by decalcification in EDTA. The obtained pieces were embedded in paraffin separately. After they were taken to the microtome, obtaining of 6μm thick sections and are mounted on slides. The blades aimed at quantitative analysis were stained with hematoxylin and eosin. To perform the analysis we used an optical microscope at 2.5x increase objective coupled to an image capture camera and connected to a computer with a software analyzer scanned images. The extent of bone formation in the defect area was measured. To this end, the use of some criteria was required. The total area of the defect in the analyzed region should match the area occupied by the bone of the calvaria in the same region before the defect production. For determination of the total area, should be set the height of the defect, having as parameter the thickness of bone at the defect edge. Once obtained the values of each blade, they are summed... |

Once obtained the values of each blade, they are summed...A atrofia óssea pode ser resultado de etiologias como patologias, traumas, consequência de procedimentos cirúrgicos e da reabsorção óssea fisiológica após a perda do elemento dentário. Este fato tem proporcionado extensos estudos sobre o processo de reparação óssea. O objetivo deste estudo foi avaliar a eficácia do uso da membrana absorvível de colágeno em defeitos ósseos críticos realizados em calvaria de ratos. Para tal 36 rattus norvegicus foram divididos em grupos controle e tratado. Os animais tiveram defeitos de 7 mm de diâmetro realizados em suas calvárias, especificamente sobre a sutura sagital mediana nos ossos frontal e parietal direito e esquerdo. No grupo controle o defeito foi preenchido apenas por coágulo sanguíneo, enquanto o grupo tratado foi preenchido por coágulo sanguíneo(GC) e recoberto por membrana absorvível (GCM) de colágeno bovino. Os animais sofreram eutanásia nos períodos de 7, 30 e 60 dias pós-operatórios. A calvaria foi removida e o material excedente foi eliminado, deixando apenas os ossos cranianos. As peças foram fixadas em formol 10%, seguidos da descalcificação em EDTA. As peças obtidas foram incluídas em parafina separadamente. Após foram levadas ao micrótomo, obtendo assim cortes de 6μm de espessura e serão montadas em lâminas. As laminas destinadas a analise quantitativa foram coradas por hematoxilina e eosina. Para realizar a analise foram utilizados um microscópio óptico com objetiva de aumento de 2,5x acoplado a uma câmera de captura de imagem e conectado a um computador, com um software analisador de imagens digitalizada. Foi mensurado a extensão da área de formação óssea nos defeitos. Para tal, foi necessário a utilização de alguns critérios. A área total do defeito na região analisada devia corresponder a área ocupada pelo osso da calvaria, na mesma região antes da confecção do defeito.
