Leucograma e perfil bioquímico sérico de cabras com mastite experimental causada por Staphylococcus aureus e Streptococcus agalactiae

Resumo: Acute phase proteins (APP) are sensitive and early indicators of inflammation. However, information regarding goats APP are scarce in the literature. So the aim of this study was to determine the leukocyte count and serum biochemical profile, in particular APP, in goats with induced Staphylococcus aureus (Group I) and Streptococcus agalactiae (Group II) mastitis in order to verify its importance as early biomarkers of mammary infection in these species. GI was composed of five Saanen goats inoculated through the teat canal with about 9.5 x 109 colony forming units (CFU) of S. aureus and GII consisted of five French Alpine goats inoculated with approximately 4.4 x 1011 CFU of S. agalactiae. The experimental moment immediately prior to inoculation was designated M0 and the following identified as M1, M2, M3, M4, M5, M6 and M7 when blood and milk samples were obtained. Leucocyte count and measurement of plasma fibrinogen concentration, serum levels of aspartate aminotransferase, alkaline phosphatase, gamma glutamyl transferase, creatine kinase, total protein, albumin, urea, creatinine, total calcium, phosphorus, magnesium and iron, were determined with blood samples, using commercial kits. Serum transferrin, ceruloplasmin, haptoglobin and α1-acid glycoprotein were obtained by the SDS-Page Electrophoresis technique. Milk samples were used to monitor the development of clinical mastitis through the strip cup test, California Mastitis Test (CMT) and microbiological analyzes. Data were subjected to analysis of variance and the means to Dunnet test, considering a 95% significance level (p < 0.05). It was noted that the experimental infection model developed for the induction of clinical mastitis in goats was effective in both groups. No entanto, as informações disponíveis na literatura em relação à espécie caprina são escassas. Assim, o objetivo deste estudo foi determinar o leucograma e o perfil bioquímico sérico, em especial de PFA, de cabras com mastite induzida por Staphylococcus aureus (Grupo I) e Streptococcus agalactiae (Grupo II), a fim de verificar sua importância como biomarcadores precoces da infecção mamária nesta espécie. O GI foi composto por cinco cabras da raça Saanen, inoculadas por via intramamária com cerca de 9,5 x 109 unidades formadoras de colônia (UFC) de S. aureus e o GII composto por cinco cabras da raça Alpina Francesa inoculadas com aproximadamente 4,4 x 1011 UFC de S. agalactiae. O momento experimental imediatamente anterior à inoculação foi denominado M0 e os seguintes identificados como M1, M2, M3, M4, M5, M6 e M7, nos quais eram realizadas colheitas de sangue e leite. Com as amostras de sangue foram obtidos o leucograma, mensuração da concentração plasmática de fibrinogênio, determinação do teor sérico de aspartato aminotransferase, fosfatase alcalina, gamaglutamiltransferase e creatina quinase, além das concentrações séricas de proteína total, albumina, ureia, creatinina, cálcio total, fósforo, magnésio e ferro, utilizando-se kits comerciais. As concentrações séricas de transferrina, ceruloplasmina, haptoglobina e α1-glicoproteína ácida foram obtidas por meio da técnica de eletroforese SDS-Page. Com as amostras de leite foi realizado acompanhamento da evolução da mastite por meio dos testes da caneca de fundo escuro, California Mastitis Test (CMT) e análises microbiológicas.
