Treinamento físico e antioxidantes enzimáticos no tecido cerebral de ratos diabéticos experimentais

Resumo: | Several studies have demonstrated different effects of physical exercise (FE) upon the antioxidant defense system. However, when it is associated with Diabetes Mellitus (DM) these effects require clarification, thus, the present study aimed to investigate the actions of a protocol of exercise on antioxidant system and other parameters in blood and brain tissue of diabetic rats . For that we used male Wistar rats divided in sedentary control (SC), trained control (TC), sedentary diabetic (SD) and trained diabetic (TD). The training session consisted of a swimming 1 hour per day for 5 consecutive days per two days of rest for 8 weeks, with a load equivalent to the anaerobic threshold of 5.8 % and of 4.2 % of the body mass for TC group and for DT group, respectively. We assessed the concentration of blood glucose, liver glycogen and of the thiobarbituric reactive substances (TBARS), the activity of the antioxidant enzymes: catalase (CAT) and superoxide dismutase (SOD) in brain and blood. Also it was quantified the expression of SOD isoforms 1 and 2 in the cerebellum. As expected, exercise acted reducing blood glucose in the TD group. Although the FE has remained oxidative standards of sedentary control group , when associated with DM, it was unable to combat lipid peroxidation in both tissues. CAT activity was increased by DM in the brain, but in blood tissue the same activity was increased due to the EF associated to the DM or not. The activity of SOD (Cu/Zn, Mn and Fe) was not significantly different between the groups, indicating that its isoforms and their mechanisms of action acted homogeneously in brain tissue. However, in the cerebellum, exercise increased the protein expression of both isoforms of SOD (1 and 2) independently of pathological action indicating an improvement in antioxidant activity by the exercise. Porém quando associado ao Diabetes Mellitus (DM) esses efeitos carecem de esclarecimentos, sendo assim o presente estudo objetivou investigar as ações de um treinamento físico sobre o sistema antioxidante e outros parâmetros no tecido cerebral e sanguíneo de ratos diabéticos. Para isto foram utilizados ratos machos Wistar divididos em 4 grupos aleatórios: controle sedentário (CS), controle treinado (CT), diabético sedentário (DS) e diabético treinado (DT). O treinamento físico consistiu de uma sessão de natação de 1 hora por dia, durante 5 dias consecutivos por dois dias de descanso, durante 8 semanas, com uma carga equivalente ao limiar anaeróbio de 5,8 % e 4,2 % da massa corporal para o grupo CT e o grupo DT, respectivamente. Avaliou-se as concentrações de glicose no sangue, glicogênio hepático e de substâncias que reagem ao ácido tiobarbitúrico (TBARS), a atividade das enzimas antioxidantes catalase (CAT) e superóxido dismutase (SOD) no cérebro e no sangue. Quantificou-se a expressão das isoformas SOD 1 e 2 no cerebelo através da técnica de Western Blotting. O treinamento físico atuou reduzindo a glicemia para o grupo DT e quando associado ao DM não foi capaz de combater a peroxidação lipídica em ambos os tecidos analisados. A atividade da CAT foi aumentada pelo DM e pelo TF no sangue e apenas pelo DM no cérebro. A atividade da SOD (Cu/Zn, Mn e Fe) não teve diferença significativa entre os grupos indicando que suas isoformas e seus respectivos mecanismos de ação atuaram de forma homogênea no tecido cerebral. Entretanto, no cerebelo, o TF elevou a expressão protéica das isoformas da SOD independentemente da ação do DM indicando aumento na ação antioxidante.
