Efeito da teriparatida sobre o processo de reparo alveolar em ratos orquiectomizados: análise imunoistoquímica e microtomográfica

Objective: to evaluate the influence of teriparatide on the alveolar bone repair in orchiectomy rats through tridimensional microtomography and immunohistochemical analysis. Materials and Methods: 54 male and adults rats were divided into three experimental groups: 18 control rats (SHAM); 18 orchiectomized rats (ORQ ST); and 18 orchiectomized rats treated with teriparatide (ORQ TERI). At 30 days after the orchiectomy the teriparatide drug treatment started in the ORQ TERI group, at 60 days, the right upper incisor of the animals of all groups was extracted and at 14 and 42 days after the extraction the rats were euthanized. For the immunohistochemical analysis, the expression of the wnt, alp, oc and trap proteins was analyzed. For the microtomography (micro CT) evaluation of volume and percentage of bone volume (BV, BV/TV), trabecular thickness (Tb.Th), number and separation of trabeculae (Tb.N, Tb.Sp). The quantitative data of the micro CT underwent the homoscedasticity analysis (Shapiro-Wilk test), to distinguish the parametric and nonparametric data. The parametric data were compared by ANOVA 1 factor test and Bonferroni post-test, with significance level p <0.05. Results: In the immunohistochemistry analysis, there was a moderate expression of wnt in the SHAM and ORQ TERI groups at 14 days, becoming intense at 42 days. Alp presented moderate expression only in ORQ TERI and light in the other groups in both periods. OC was similar in all groups and periods with light expression. The trap presented moderate expression in SHAM and ORQ ST groups and light in ORQ TERI group at 14 and 42 days. For the BV parameter, the ORQ TERI presented the highest value (157.91mm³) in relation to the other groups (ORQ ST: p < 0.05).
