Resumo: | Aphids are the main pests of winter cereals. In addition to direct damage by sap suction, they are vectors of Barley yellow dwarf virus and Cereal yellow dwarf virus (B / CYDV). It is necessary to enable the sustainable management of this pathosystem by improving population monitoring methods and decision-making criteria (action levels), estimate the damage to production caused by B / CYDV epidemics and determine the effectiveness and profitability of insecticide management to control the problem over time. The aim of this study was to broaden the understanding of the aphid population dynamics and to evaluate insecticide management alternatives to support the management of the aphid-virose complex of yellow dwarfism in wheat crop, in Passo Fundo/RS region. In the 2013/14/15 and 16 wheat crop seasons, the aphid population was monitored using a yellow - tray trap with water and direct counting on plants in the control plots, in which five systems were evaluated: Insecticide management: TT - total treatment to evaluate the productive potential (insecticide seed treatment + weekly insecticide spraying on the shoot); TS - seed treatment only; PA - only insecticides in shoots when reaching action level (NA); TS + PA and SI - no insecticides (witness). In 2013, the trial was not treated with TS + PA. Generally, when below NA, the number of aphids in plants and trays fluctuated proportionally. After reaching NA, the aphid population increased significantly in both evaluations (plants and trays). In the average of the 2013/14/15 and 16 harvests, in the TT, a yield of 4,573.95 kg/ha was obtained. Considering this treatment as a reference, the second treatment with higher yield was TS + PA (4,121.26 kg / ha), followed by PA (3,946.69 kg/ha) and TS (3,921.62 kg/ha). At the other extreme, SI provided 3,626.05 kg ha. The TS + PA treatment presented the best cost x benefit ratio, with a net revenue of R$ 287.49 h -1, compared to the control (SI). |

The TS + PA treatment presented the best cost x benefit ratio, with a net revenue of R$ 287.49 h -1, compared to the control (SI).Para estabelecer a técnica de transformação genética de plantas via Agrobacterium tumefaciens, um dos pontos críticos é ajustar os fatores que influenciam a transformação, tais como genótipo da planta e condições de infecção e cocultivo dos explantes, aplicando estratégias que possam aumentar as frequências de transferência do T-DNA como aplicação de estresses antes e durante a infecção dos explantes com a agrobactéria e ajuste da concentração de compostos fenólicos nos meios de infecção e cocultivo. Dessa forma, este trabalho objetivou adequar um protocolo de transformação genética de milho via A. tumefaciens para dois genótipos de milho. O primeiro capítulo deste trabalho avalia o efeito da submissaõ dos embriões imaturos do hibrido Hi-II e da variedade brasileira BR 451 a diferentes condições infecção com a A. tumefaciens EHA101:pTF101, a qual carrega os genes bar e uidA sob controle do promotor CaMV35S. No primeiro experimento os embriões foram submetidos a diferentes temperaturas de infecção (43 oC e temperatura ambiente). No segundo experimento foi aplicado um estresse combinado (46 oC por 3 minutos, resfriamento em gelo por 1 minuto, centrifugação a 12.000 rpm a 4 oC por 10 minutos) antes da infecção com A. Os afídeos são as principais pragas de cereais de inverno. Além de danos diretos pela sucção de seiva, são vetores de Barley yellow dwarf virus e Cereal yellow dwarf virus (B/CYDV). Aperfeiçoar métodos de monitoramento populacional e os critérios para tomada de decisões (níveis de ação), estimar os danos à produção causados por epidemias de B/CYDV e determinar a eficácia e a rentabilidade do manejo de inseticidas, visando o controle do problema, ao longo de safras de trigo, é necessário para viabilizar o manejo sustentável desse patossistema. O objetivo deste estudo foi ampliar a compreensão sobre a dinâmica populacional de afídeos e avaliar alternativas de manejo com inseticidas para dar suporte ao manejo do complexo afídeos-virose do nanismo-amarelo na cultura do trigo, na região de Passo Fundo/RS. Nas safras tritícolas 2013/14/15 e 16, a população de afídeos foi acompanhada com o auxílio de armadilha do tipo bandeja - amarela com água e contagem direta em plantas nas parcelas testemunhas de experimento de controle da praga, no qual foram avaliados cinco sistemas de manejo com inseticidas: TT - tratamento total, para avaliar o potencial produtivo (tratamento de sementes com inseticida + pulverização semanal de inseticidas na parte aérea); TS - somente tratamento de sementes; PA - somente inseticidas em parte aérea ao atingir o nível de ação (NA); TS+PA e SI - sem inseticidas (testemunha). No ano de 2013, o ensaio não teve o tratamento TS+PA. De maneira geral, quando abaixo do NA, o número de afídeos em plantas e em bandejas, flutuou proporcionalmente. Depois de atingido o NA, a população de afídeos aumentou significativamente em ambas as avaliações (plantas e bandejas). Na média das safras 2013/14/15 e 16, no TT obteve-se produtividade de 4.573,95 kg/ha. Considerando este tratamento como referencial, o segundo tratamento com maior produtividade foi TS+PA (4.121,26 kg/ha), seguido de PA (3.946,69 kg/ha) e TS (3.921,62 kg/ha). No outro extremo, o SI proporcionou 3.626,05 kg/ha.
