Resumo: In this work we evaluated the immune response of silver catfish (Rhamdia quelen) following the inoculation of different combinations of antigens and adjuvants. In chapter 1 we investigated the potential of inactivated Parapoxvirus ovis (iPPVO) as immunomodulator of the innate immune response and production of antibodies to bovine serum albumin (BSA). The respiratory burst of silver catfish juvenile (60 80 g) inoculated with iPPVO (1 x 106 plaque forming units PFU) was similar, at 24 h post-inoculation (p.i.), to the control fish inoculated with sterile phosphate buffered saline (PBS) solution. However, significant differences (p < 0.05) were observed on circulating monocytes, heterophils and thrombocytes at 3, 5 and 7 days p.i., respectively in iPPVO inoculated silver catfish. Furthermore, silver catfish inoculated with iPPVO+BSA (200 &#956;g/fish) had significantly (p < 0.05) higher antibody titers to BSA compared to fish inoculated with BSA+PBS, indicating its adjuvanticity effect on antibody production. In chapter 2 we report the use of different combinations of traditional adjuvants (aluminum hydroxide, Freund complete adjuvant, Freund incomplete adjuvant, and montanide), and new generation adjuvants, composed of olygodeoxynucleotides CpG (ODN CpGs) associated with Aeromonas hydrophila bacterin, or BSA, on the production of specific antibodies. In these experiments, juvenile catfish (60 80 g) were inoculated once with 0,2 ml of each antigen (A. hydrophila, 1 x 108 colony forming unit CFU, or BSA 200 &#956;g) and the production of antibodies was determined at 42 days p.i. in the groups inoculated with A. hydrophila, and at 14, 28 and 42 days p.i. for the groups inoculated with BSA. Control groups consisted of fish inoculated with the antigens and sterile PBS. All immunized fish seroconverted to A. hydrophila; however, there was a significant difference (p <0.05) between the control group and the remaining inoculated groups. And, amongst the A. hydrophila inoculated groups there was a significantly lower (p < 0.05) antibody titer in the group of fish inoculated with A. hydrophila + ODN CpG 1668 compared to the groups inoculated with A. hydrophila + CpG 2133, CpG 2102 and CpG 2143. Fish inoculated with BSA combined with different adjuvants, all fish seroconverted and there were significant differences (p < 0.05) in the antibody titers at 14, 28 and 42 days p.i. The difference in antibody titers were observed either in fish within the same group at different days p.i., or when compared within different groups in the same day p.i. In general, higher antibody titers to BSA were observed at 28 days p.i. Within the classical adjuvants, the higher anti-BSA antibody titers were observed in fish inoculated with a combination of BSA+Freund complete adjuvant. And, within the ODN CpGs adjuvants, higher anti-BSA antibody titers were observed in fish inoculated with BSA+CpG 1668. The studies reported herein indicate that iPPVO might be exploited as immunomodulator in fish and that ODN CpGs adjuvants stimulated the production of specific antibodies in silver catfish at titers similar or even higher than those stimulated by classical adjuvants

No capitulo 1 investigou-se o uso do Parapoxvirus ovis inativado (iPPVO) como modulador do sistema imune inato e na inducao de anticorpos anti-albumina serica bovina (BSA). Juvenis de jundias (60 - 80 g) inoculados com iPPVO (1x106 unidades formadoras de placas - UFP) nao apresentaram alteracoes significativas na explosao respiratoria 24 h apos a inoculacao quando comparados com jundias inoculados com solucao salina fosfatada (PBS) esteril. No entanto, observaram-se alteracoes significativas (p < 0.05) na populacao de monocitos, heterofilos e trombocitos aos 3, 5 e 7 dias pos-inoculacao (p.i.), respectivamente. Alem disso, o uso do iPPVO como adjuvante da BSA (200 Êg/peixe) induziu titulos de anticorpos significativamente maiores daqueles observados em jundias imunizados somente com BSA, indicando um efeito adjuvante na inducao de anticorpos especificos. O capitulo 2 relata o uso de diferentes combinacoes de adjuvantes tradicionais (hidroxido de aluminio, adjuvante completo de Freund, adjuvante incompleto de Freund e Montanide) e adjuvantes de nova geracao, constituido por oligodeoxinucleotideos CpG (ODN CpGs ) associados a bacterina de Aeromonas hydrophila, ou BSA, na inducao de anticorpos especificos. Nesses experimentos, juvenis de jundias (60 - 80 g) foram inoculados uma unica vez com 0,2 ml de cada combinacao de antigeno (A. hydrophila = 1x108 unidades formadoras de colonias - UFC; BSA 200 Êg) e a presenca de anticorpos especificos foi determinada aos 42 dias p.i. para o grupo inoculado com A. hydrophila, e aos 14, 28 e 42 dias p.i. para o grupo inoculado com BSA. Grupos controles constituiram-se de peixes inoculados com os respectivos antigenos PBS esteril. Todos os peixes imunizados soroconverteram a inoculacao com A. hydrophila, no entanto houve uma diferenca significativa entre o grupo controle (A. hydrophila+PBS) e os demais grupos inoculados. E, entre os grupos inoculados com A. hydrophila e diferentes combinacoes de adjuvantes, houve uma diferenca significativamente menor (p < 0.05) no titulo de anticorpos do grupo de peixes inoculado com A. hydrophila + ODN CpG 1688 em relacao aos grupos inoculados com A. hydrophila + ODN CpG 2133, CpG 2102 e CpG 2143. Nos peixes inoculados com as diferentes combinacoes de adjuvantes e BSA, todos os peixes soroconverteram, e houve diferencas significativas (p < 0.05) nos titulos de anticorpos detectados aos 14, 28 e 42 dias p.i. Essas diferencas foram observadas tanto em relacao aos peixes do mesmo grupo em diferentes dias p.i., como entre diferentes grupos de peixes quando comparados no mesmo dia p.i. De forma geral, os maiores titulos de anticorpos foram observados aos 28 dias p.i. E, entre os adjuvantes classicos, os titulos significativamente maiores de anticorpos anti-BSA foram observados com o uso do adjuvante completo de Freund. E, entre os adjuvantes ODN CpGs, os maiores titulos de anticorpos anti-BSA foram observados com o uso do ODN CpG 1668. Os estudos descritos no presente trabalho indicam que o iPPVO pode ser explorado como imunomodulador em peixes e que adjuvantes ODN CpGs induziram anticorpos especificos anti-BSA em jundias em niveis similares ou superiores aos adjuvantes classicosMade available in DSpace on 2018-01-10T17:52:52Z (GMT).
