Resumo: | The canine mammary tumors has high incidence and malignancy being provoked by several risk factors including age, hormonal activity, nutrition, virus, pseudopregnancy and exogenous progestagen. In this work the goal was to characterize the canine mammary tumors, evaluate the implication of the adjacent mamma and the risk factors involvement on the occurrence of the tumors. They were used 50 bitches with mamma tumor diagnosis and submitted to mastectomy of the mamma and 11 normal bitches that were submitted for elective ovariosalpingohisterectomy in Hospital Veterinary of UFRPE. They were picked biopsies of the tumors and of the normal mammas for histopathologic analysis according to the routine techniques. The animals went of varied races having age comprehended between 4 and 19 years for the bitches with tumor and between 7 months and 6 years for the normal bitches, and the ages between 9 and 13 years were to what it presented the biggest neoplasic frequency (48%). Of the 50 tumors, 68% were carcinosarcoms, 24% were carcinomas, 2% benign mixed tumor, 2% hemorrhagic, 2% fibrosises and 2% were inflammatory processes. The relation of the tumors with the risk factors did not are demonstrate when compared between the malignant tumors and between the normal bitches and the tumor carriers. Of the adjacent mammas, just 36% were normal, 48% presented carcinoma, 6% carcinosarcoms, 2% hiperplasy, 2% fibrosis and in 6% the material was not enough to guarantee the diagnosis. It concludes that in these animals there was no interference of the factors pseudopregnancy and contraceptives use in the appearance of the tumors, but that the age is an important factor, being the more seniorest susceptible. Considering the tumors high frequency in the adjacent mammas to tumour mammas, it suggests the withdrawal of all the mammary chain of the attacked side |

O gene p53, conhecido como um gene supressor de tumor, tem apresentado mutações relacionadas com neoplasias. Neste trabalho, o objetivo foi caracterizar os tumores mamários em cadelas, avaliar o comprometimento da mama lateral ao tumor e o envolvimento de fatores de risco sobre a ocorrência dos tumores; analisar o gene p53, na região entre os exons 4 e 8 e relacionar os tumores malignos e mamas normais com as mutações ocorridas neste gene. Foram utilizadas 50 cadelas com tumor de mama e 11 cadelas normais que foram atendidas no Hospital Veterinário da Universidade Federal Rural de Pernambuco. Foram colhidas amostras dos tumores e das mamas adjacentes, clinicamente normais, bem como biopsias de mamas das cadelas normais, para análise histopatológica segundo as técnicas rotineiras. Outros fragmentos foram processados para extração do DNA, amplificação dos exons pela técnica de reação em cadeia da polimerase (PCR) com oligonucleotídeos específicos e sequenciamento. Os animais foram de raças variadas tendo idade compreendida entre 4 e 19 anos para as cadelas com tumor e entre 7 meses e 6 anos para as cadelas normais, sendo que a faixa etária entre 9 e 13 anos foi a que apresentou a maior freqüência de neoplasias (48%). Das 50 alterações mamárias, 68% foram carcinosarcomas, 24% foram carcinomas, 2% tumor misto benigno, 2 hemorrágicas, 2% fibroses e 2% foram processos inflamatórios inespecíficos. Não houve diferença na relação entre os fatores de risco e a ocorrência de tumores nas cadelas. Das mamas laterais aos tumores, apenas 36% foram normais, 48% apresentavam carcinoma, 6% carcinosarcomas, 2% hiperplasia, 2% fibrose e em 6% o material não foi suficiente para garantir o diagnóstico. Foram escolhidas amostras de 9 carcinomas e 7 carcinosarcomas com as respectivas mamas normais adjacentes, e de 6 cadelas normais, que foram seqüenciadas em seqüenciador automático e analisadas para a presença de mutações em comparação com a seqüência de nucleotídeos da p53 canina (GenBank - S77819). Foram encontradas mutações em 85% das amostras que após o sequenciamento apresentaram homologia com a proteína p53 canina, de 86,3% em média (GenBank - AAB42022.1). Das mutações observadas, as mais freqüentes foram as missense e os exons mais acometidos foram o 5 e o 8 com, respectivamente, 23,2% e 24,9% das mutações. As alterações de nucleotídeos mais freqüentes foram deleções e inserções, que resultaram na maioria das mutações missense. Conclui-se que nos animais estudados não houve interferência dos fatores pseudogestação e uso de anticoncepcionais no surgimento dos tumores, mas que a idade é um fator relevante, sendo, entretanto, a idade avançada considerada um fator de risco elevado. Considerando a alta freqüência de tumores nas mamas laterais às neoplásicas e a ocorrência de mutações nos genes destas amostras, sugere-se que na exerése do tumor seja retirada a cadeia lateral completa para evitar recidiva.
