Resumo: | The contagious agalactia of ovine and caprine (CAOC) is characterized by a decline and subsequent failure of milk production, arthritis and keratoconjunctivitis causing economic losses in dairy goats and sheep herds. Classic diagnostic is performed by culture and identification of mycoplasmas species. The utilization only of this method to diagnostic has contribute for dissemination of infection and reduced efficiency of the controls measures because your necessity of long time to the diagnostic conclusion. The objective this study was performed and standardized an indirect ELISA using conjugate with protein-G (ELISA-G) and two types of obtained by growth of M. agalactiae antigen. Whole bacterial and sonicated culture was used as antigen to sensibility on microtitre plates. A total of 46 serum sample were obtained in infected goat herd with clinical signs in forty-five period and 40 samples in free herds of disease to standardization and application of ELISA test. The infection status was confirmed by culture of M. agalactiae from milk. Do not had statistical difference between positive and negative controls serum (P/N) when whole or sonicated antigen wereused. The sensibility of ELISA-Gt and ELISA-Gs were of 77,27% and 88,63%, respectively. Specificity were 95,24% for both ELISA. In conclusions, the indirect ELISA-Gt or ELISA-Gs should be utilized as tools for diagnostic of contagious agalactia in goats. |

O diagnóstico clássico é feito pelo cultivo e isolamento dos micoplasmas envolvidos. A utilização dessa técnica como único método diagnóstico, contribui para a disseminação da infecção e dificulta a implantação de medidas eficazes de controle por requerer longo tempo para conclusão diagnóstica. Por isso, o objetivo deste trabalho foi desenvolver e padronizar um ELISA indireto utilizando a proteína G (ELISA-G) como conjugado e aplicação de dois tipos de antígenos produzidos a partir de M. agalactiae. Antígeno total e sonicado foram utilizados para sensibilizar as microplacas. A padronização da técnica foi realizada utilizando soros caprinos com e sem sinais clínicos de agalaxia contagiosa. Quarenta e seis soros caprinos coletados com intervalo de 45 dias, obtidos em rebanho infectado com sinais clínicos e 40 soros caprinos de rebanhos livres da infecção foram utilizados para avaliação da técnica. A presença de M. agalactiae no rebanho foi confirmada por cultivo de leite em meio Hayflick modificado. Não houve diferença significativa entre os valores da razão positivo/negativo (P/N) dos soros controles quando utilizado antígeno total ou sonicado. A sensibilidade relativa do ELISA-Gt e ELISA-Gs foi de 77,27% e 88,63%, respectivamente, enquanto que a especificidade foi de 95,24% para ambos. Pesquisa de anticorpos em rebanho com quadro clínico característico revelou aumento significativo dos percentuais de densidade óptica (DO) em soros caprinos, coletados com intervalo de 45 dias.
