Resumo: | The aim of this study was to evaluate the in vitro antimicrobial effects of pure White Portland cement and zirconium oxide plus in the ratio of 1:1 (50% white Portland cement and 50% zirconium oxide), added amoxicillin and ciprofloxacin after solidification and also determine the minimum inhibitory concentration and minimum lethal concentration of these products. After adding up to 10% of cement weight of amoxycillin and ciprofloxacin, separately, they were spatulate with distilled water, awaiting prey 24 hours at 37 ° C. The material was crushed manually in degree and pestle until the consistency of fine powder and smooth. One hundred and twenty vials were prepared containing BHI and cements were inserted directly into the tubes at concentrations of 5 mg / ml, 2.5 mg / ml, 1.25 mg / ml, 0.625 mg / ml and 0.313 mg / ml and mechanically agitated for better dispersion. Strains from the American Type Culture Collection (ATCC) of Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa (ATCC 27853), Escherichia coli (ATCC 25922) and Enterococcus faecalis (ATCC 29212) were obtained. A fresh inoculum of each organism was prepared and seeded directly in the tubes, each tube in each subgroup received a microorganism at each concentration. The tubes were then incubated at 37 ° C and the turbidity was assayed at 24 and 48 hours. After 24 hours incubation at 37 ° C, all tubes of the groups Portland Portland 50% pure and were hazy, indicating growth microbiano.No subgroup Portland + amoxicillin MIC was only Escherichia coli concentration of 2.5 mg / ml and 5 mg / ml. Após adicionar-se aos cimentos 10 % de seu peso de amoxicilina e ciprofloxacina, separadamente, os mesmos foram espatulados com água destilada, aguardando-se a presa de 24 horas, em estufa a 37°C. O material obtido foi triturado manualmente em grau e pistilo até a consistência de pó fino e homogêneo. Cento e vinte tubos de ensaio contendo BHI foram preparados e os cimentos foram inseridos diretamente nos tubos, nas concentrações de 5 mg/ml, 2,5 mg/ml, 1,25 mg/ml, 0,625 mg/ml e 0,313 mg/ml, e agitados mecanicamente, para melhor dispersão. Cepas da American Type Culture Collection (ATCC) de: Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa (ATCC 27853), Escherichia coli (ATCC 25922) e Enterococcus faecalis (ATCC 29212), foram obtidas. Um inoculado fresco de cada microrganismo foi preparado e semeado diretamente nos tubos, sendo que cada tubo em cada subgrupo recebeu um microrganismo em cada concentração. Os tubos foram, então, incubados em estufa a 37°C e a turbidez foi avaliada nos períodos de 24 e 48 horas. Após 24 horas de incubação em estufa a 37°C, todos os tubos dos grupos Portland puro e Portland 50% estavam turvos, indicando crescimento microbiano. No subgrupo Portland+amoxicilina, houve CIM somente para Escherichia coli na concentração de 2,5 mg/ml e 5 mg/ml. No subgrupo Portland 50%+amoxicilina, houve CIM somente para Escherichia coli, na concentração de 5 mg/ml. Em todas as outras concentrações, ocorreu crescimento bacteriano. Os subgrupos Portland+ciprofloxacina e Portland 50%+ciprofloxacina, não apresentaram crescimento bacteriano em todas as concentrações. Em todos os controles positivos, ocorreu crescimento bacteriano. Concluímos que acréscimo de ciprofloxacina ao cimento Portland favoreceu o efeito inibitório e bactericida ao cimento após a presa, in vitro, e a amoxicilina, apenas na concentração de 2.5 μg/ml para Portland puro e 5 μg/ml para Portland 50%, favoreceram o efeito inibitório para E. coli.Made available in DSpace on 2015-10-20T18:56:10Z (GMT).
