Mechanisms Involved in the Regeneration of Peripheral Nerve Injuries

Resumo: | Peripheral nerve injuries are common and result in motor sensory and autonomic disability. Even in the best circumstances regeneration is rarely completely effective. The peripheral nerve regeneration involves the integration of various cellular and molecular processes followed by a series of changes observed in structures related to the injury site. In recent years the knowledge on mechanisms involved in this process has been deeply studied through clinical and experimental research. The expectation of inclusion of new treatment strategies, including neuro-engineering, molecular therapy, neuroprotective pharmacotherapy, modulation of neurotrophic factors, transplanting of embryonic cells and recent knowledge on brain plasticity secondary to these injuries may be added to current concepts of surgical and conservatory treatment for better functional results. Current study presents the main mechanisms involved in injury and regeneration of peripheral nerves and the perspectives of new treatment strategies. |

A regeneração do nervo periférico envolve a integração de diversos processos celulares e moleculares que se seguem em uma série de modificações observadas nas estruturas relacionadas ao local da lesão. Nos últimos anos o conhecimento dos mecanismos envolvidos neste processo tem sido aprofundado por meio de pesquisas clínicas e experimentais. A expectativa da inclusão de novas estratégias de tratamento, incluindo a neuroengenharia, terapia molecular, farmacoterapia neuroprotetora, modulação de fatores neurotróficos, transplantes de células embrionárias e o aprofundamento dos conhecimentos sobre a plasticidade cerebral secundária a estas lesões, se soma aos atuais conceitos de tratamento cirúrgico e conservador, em busca de melhores resultados funcionais.
