Salvador, BA, BrasilBackground:ThecurrenttreatmentofChagasdisease,endemicinLatinAmericaandemerginginseveralcountries,islimitedbythefrequentsideeffectsandvariableefficacyofbenznidazole.Naturalproductsareanimportantsourceforthesearchfornewdrugs.Aim/hypothesis:Consideringthegreatpotentialofnaturalproductsasantiparasiticagents,weinvestigatedtheanti-TrypanosomacruziactivityofaconcentratedethanolicextractofPhysalisangulata(EEPA).Methods:Cytotoxicitytomammaliancellswasdeterminedusingmouseperitonealmacrophages.Thean-tiparasiticactivitywasevaluatedagainstaxenicepimastigoteandbloodstreamtrypomastigoteformsofT.cruzi,andagainstamastigoteformsusingT.cruzi-infectedmacrophages.Celldeathmechanismwasde-terminedintrypomastigotesbyflowcytometryanalysisafterannexinVandpropidiumiodidestaining.TheefficacyofEEPAwasexaminedinvivoinanacutemodelofinfectionbymonitoringbloodparasitaemiaandsurvivalrate30daysaftertreatment.TheeffectagainsttrypomastigotesofEEPAandbenznidazoleactingincombinationwasevaluated.Results:EEPAeffectivelyinhibitstheepimastigotegrowth(IC502.9±0.1μM)andreducesbloodstreamtry-pomastigoteviability(EC501.7±0.5μM).Itcausesparasitecelldeathbynecrosis.EEPAimpairsparasiteinfectivityaswellasamastigotedevelopmentinconcentrationsnoncytotoxictomammaliancells.Inmiceacutely-infectedwithT.cruzi,EEPAreducedthebloodparasitaemiain72.7%.Whencombinedwithbenznida-zole,EEPAshowedasynergisticanti-T.cruziactivity,displayingCIvaluesof0.8±0.07atEC50and0.83±0.1atEC90.Conclusion:EEPAhasantiparasiticactivityagainstT.cruzi,causingcelldeathbynecrosisandshowingsyner-gisticactivitywithbenznidazole.ThesefindingswerereinforcedbytheobservedefficacyofEEPAinreducingparasiteloadinT.cruzi-mice.Therefore,thisrepresentsanimportantsourceofantiparasiticnaturaChagas diseaseTrypanosoma cruziPhysalis angulataPlant extractDrug combinationinfo:eu-repo/semantics/openAccessreponame:Repositório Institucional da FIOCRUZ (ARCA)instname:Fundação Oswaldo Cruz (FIOCRUZ)instacron:FIOCRUZLICENSElicense.txtlicense.txttext/plain;
