Resumo: | In the present work sludge samples were collected from two domestic wastewater treatment stations of Sanepar, Curitiba-PR. One sample was taken from the anaerobic lagoon (LA) and the other from the fluidized bed anaerobic reactor (RALF). Eight procedures were employed for DNA extraction from the communities present in the collected samples, and two of them (extraction procedures 1 and 2) were effective in obtaining a quality product. After obtaining DNA of quality, for both samples LA and RALF, amplifications of target fragments encoding the 16S rDNA region of Archaea were made. Amplifications were performed by PCR using the primers PRA46f and PREA1100r, followed by Nested PCR using the primers PARCH340f and PARCH519r. Amplification conditions were modified many times in order to achieve higher quality and specificity of the final product. After amplification of the fragments of around 180 bp by Nested PCR, these were purified and submitted to sequencing. The fragments were also submitted to denaturing gradient gel electrophoresis (DGGE), a technique that generated profiles of the communities present in both systems. In this way, the comparison of these profiles was possible through the principal component analysis (PCA), based on the Bray-Curtis method, allowing to verify the difference in the profile obtained for the samples RALF and LA, that indicates the difference in composition of the communities present in both systems. The similarity degree was of 65% for the communities existent in the different environments. |

The similarity degree was of 65% for the communities existent in the different environments.No presente trabalho, foram coletadas amostras de lodo provenientes de duas estações de tratamento de efluentes domésticos da Sanepar, Curitiba – PR. Sendo uma proveniente de lagoa anaeróbia (LA) e outra proveniente de reator anaeróbio de leito fluidizado (RALF). Oito procedimentos foram empregados para a extração do DNA das comunidades presentes nas amostras coletadas, sendo que dois deles (procedimentos 1 e 2 de extração) foram eficientes na obtenção de um produto de qualidade. Após a obtenção de DNA de qualidade, tanto para amostra de RALF quanto para amostra de LA, foram realizadas as amplificações de fragmentos alvo que codificam a região do 16S rDNA de Archaea. As amplificações foram realizadas por PCR utilizando os iniciadores PRA46f e PREA1100r, seguidas de Nested PCR utilizando os iniciadores PARCH340f e PARCH519r. As condições de amplificação foram modificadas diversas vezes a fim de atingir uma maior qualidade e especificidade do produto final. Após a amplificação do fragmento de aproximadamente 180 pb pela Nested PCR, os mesmos foram purificados e submetidos a sequenciamento. Os fragmentos foram também submetidos à técnica de eletroforese em gel de gradiente desnaturante (DGGE) que gerou perfis das comunidades presentes em ambos os sistemas. Dessa forma, foi possível a comparação desses perfis e através da análise de componente principal (PCA), com base no método de Bray-Curtis, e assim verificar a diferença no perfil obtido para as amostras de RALF e de LA, que indica a diferença na composição das comunidades presesntes nos dois sistemas.
