Avaliação da imunoexpressão da osteoprotegerina, RANKL e IP3R no reparo ósseo craniofacial tratados com autoenxerto na presença de alendronato em ratas uterosalpingo-ovariectomizadas

Resumo: | Bisphosphonates are drugs known to be modulators of bone remodeling, with alendronate as its most widely used agent. Its mode of action is the modulation of the mevalonate-cholesterol pathway, being the inhibitor of osteoclastogenesis and favorable to osteogenesis. The objective of this study was to evaluate the immunoexpresion of the IP3R protein, as well as the analysis between OPG / RANKL in critical defects filled with particulate autogenous bone, besides the quantification of the bone matrix present in the defects in 60 ovariectomized utero-salpingo adult rats in the presence or absence of administration of 1mg / kg of sodium alendronate. After 15 and 60 days, euthanasia and histological, histomorphometric and immunohistochemical results were performed. The results obtained demonstrated a greater bone quantification of the Alendronate group in 60 days. The Rank-L showed greater expressiveness to non-control group, especially in 15 days, with the value increasing based on bone matrix deposition. The OPG AND IP3R, unlike Rank-L, presented themselves noticeably higher in the specimens treated with Alendronate, more specifically in the peripheral areas of the pattern and areas surrounding the autograft. The results coincided with increased bone deposition in specimens receiving alendronate. Within the limitations of this study, it was concluded that the use of Alendronate increases osteoconductivity in craniofacial defects, in addition to increasing the reference level between OPG / RANKL, besides the greater expression of IP3R, an important regulator of OPG. |

Tem como modo de ação a modulação da via mevalonato-colesterol, sendo inibidor da osteoclastogenense e favoravel à osteogênese. O objetivo deste estudo é avaliar a imunoexpressão da proteína IP3R, bem como a proporção entre OPG/RANKL em defeitos críticos preenchidos com osso autógeno particulado, além da quantificação de matriz óssea presente nos defeitos em 60 ratas adultas útero-salpingo ovariectomizados, na presença ou ausência da administração de 1mg/kg de Alendronato de sódio. Após 15 e 60 dias, foram realizadas as eutanásias e obtidos os resultados histológicos, histomorfométricos e imunohistoquimicos. Os resultados obtidos demonstraram uma maior quantificação óssea do grupo Alendronato em 60 dias. O RANK-L apresentou maior expressividade no grupo controle, especialmente em 15 dias, com aumento nos valores conforme deposição de matriz óssea. Ao contrário do RANK-L, a OPG E IP3R mostraram-se nitidamente maior em espécimes que receberam Alendronato, expressando-se principalmente em áreas periféricas do defeito ósseo e áreas circunjacentes ao autoenxerto. Estes resultados coincidiram com maior deposição óssea nos espécimes que receberam alendronato.
