Avaliação do grau de penetração intratubular de diferentes cimentos endodônticos por meio de microscopia confocal a laser

Resumo: The aim of this study was to evaluate the intratubular penetration of three bioceramic sealers – Endo Sequence, Bio-C Sealer and Sealer Plus BC – and one epoxy sealer – AH Plus –, using confocal laser scanning microscopy (CLSM) and Fluo-3 dye. Sixty canals from mandibular canines were prepared and divided into four groups (n = 15): Endo Sequence, Bio-C Sealer and Sealer Plus BC, and AH Plus. Fluo-3 dye was added to each sealer before processing. Afterwards, three 1 mm thick discs were obtained from the apical, middle, and cervical thirds of each sample and analyzed using CLSM to determine the area (mm2) and maximum penetration depth (μm) of the sealers. The Photoshop CC V.19.1.2 program was used to determine the sealer penetration area (mm2) and the ImageJ® V1.46a software was used to measure the maximum depth penetration of the sealers (μm). The Friedman statistical tests followed by Dunn's test were applied to verify the penetration of sealer between the root thirds within the same group and for the intergroup evaluation the Kruskal-Wallis statistical tests followed by Dunn's test (p<0.05) were applied. Endo Sequence was the only sealer that obtained similar outcome in all thirds in penetration area and depth. In the cervical and middle thirds, AH Plus presented greater depth of penetration in relation to the other groups and in the apical third there was no difference in penetration depth when compared to the other sealers. It can be concluded that, in the cervical and middle thirds, the bioceramic sealers penetrated less into the dentinal tubules than the AH Plus and, in the apical third, the penetration pattern was similar for all cements.

Sessenta canais radiculares de caninos inferiores foram preparados e divididos em quatro grupos (n = 15): Endo Sequence, Bio-C Sealer e Sealer Plus BC, e AH Plus. O corante Fluo-3 foi adicionado a cada cimento antes da obturação. Posteriormente, três discos de 1 mm de espessura foram obtidos de cada um dos terços, apical, médio e cervical. Foram obtidas imagens de cada amostra com MCVL. O programa Photoshop CC V.19.1.2 foi utilizado para determinar a área de penetração do cimento (mm2) e o software ImageJ® V1.46a foi utilizado para medir a profundidade máxima de penetração dos cimentos (μm). Os testes estatísticos Friedman seguido do teste de Dunn foi utilizado para verificar a penetração do cimento entre os terços radiculares dentro do mesmo grupo e para a avaliação inter grupos foram utilizados os testes estatísticos Kruskal-Wallis seguido pelo teste de Dunn (p<0,05). O Endo Sequence foi o único cimento que obteve resultados semelhantes em todos os terços em área de penetração e profundidade. Nos terços cervical e médio, o AH Plus apresentou maior profundidade de penetração em relação aos demais grupos e no terço apical não houve diferença de profundidade de penetração quando comparado aos outros cimentos.
