Seedling regeneration of Eucalyptus grandis by in vitro direct organogenesis

Resumo: Eucalyptus grandis is one of the most cultivated forestry species in Brazil due to its fast growth andgood timber quality. With the objective of cloning E. grandis for seedling production, this study aimed to improve the micropropagation protocol by direct organogenesis. Micro cuttings from in vitro germinated seedlings wereinoculated in culture medium with different levels of growth regulators during the multiplication and rooting stages. During the multiplication stage, the addition of growth regulators did not promote significant increase inbud number and height when compared to the control. Addition of ANA and IBA to the medium promoted higher rate of rooting and growth of roots, but an increase in concentration from 0,5 to 1mg/L of these growth regulatorsreduced the root growth. The results of this study evidenced the possibility of micropropagation of Eucalyptus grandis by in vitro direct organogenesis.

O Eucalyptus grandisé a espécie florestal que está entre as mais cultivadas no Brasil devido a seu rápido crescimento e por apresentar alta qualidade da madeira. Com o objetivo de clonar o E. grandis para a produção de mudas, este trabalho propôs melhorar o protocolo de micropropagação por organogênese direta. Segmentos nodais das plântulas germinadas in vitro foram inoculados em meio de cultura com diferentes níveis de reguladores de crescimento na fase de proliferação e enraizamento. Na fase de proliferação, a adição de reguladores de crescimento não promoveu aumento significativo quanto ao número e à altura de brotos em comparação ao meio de cultura ausente de reguladores. O efeito da adição de ANA e AIB ao meio de culturapromoveu elevada taxa de enraizamento e crescimento das raízes, porém o aumento da concentração de 0,5 para 1mg/L destes reguladores reduziu o crescimento das raízes. Os resultados deste trabalho evidenciam a possibilidade de micropropagação de Eucalyptus grandis a partir de organogênese direta.
